Project description:In this work, we discussed the effects Acanthopanax senticosus Harms (ASE) combined with Gastrodia elata Blume (GEB) in the treatment of CIR injury. In pharmacodynamic studies, found that co-administration of ASE and GEB may improve neuronal injury and prevent neuronal apoptosis by reducing oxidative stress and inflammation, and also help prevent CIR injury. On the basis of our hypothesis, we combined the results of transcriptomic and metabonomic analyses and found that ASE and GEB could prevent cerebral ischemia-reperfusion injury induced by middle cerebral artery occlusion by targeting phenylalanine, pyrimidine, methionine, and sphingolipid metabolism. This study provides the basis for the compatibility and efficacy of ASE and GEB.

Project description:The effects of current treatment strategies used in ischemic stroke are weakened by cerebral ischemia-reperfusion (CIR) injury. Suitable treatment regimens targeting CIR injury are still lacking. Two herbs, namely, Acanthopanax senticosus (Rupr. & Maxim.) Harms (ASE) and Gastrodia elata Blume (GEB), have been used as traditional Chinese medicine and are indicated in the treatment of stroke and cerebrovascular diseases. However, there are no studies that report the effects of ASE combined with GEB in the treatment of CIR injury. In this study, we used the Zea Longa method to induce CIR injury in male Wistar rats. Results of the pharmacodynamic studies revealed that co-administration of ASE and GEB may improve neuronal injury and prevent neuronal apoptosis by reducing oxidative stress and inflammation, and also help prevent CIR injury. On the basis of our hypothesis, we combined the results from transcriptomic and metabonomic analyses and found that ASE and GEB could prevent CIR injury by targeting phenylalanine, pyrimidine, methionine, and sphingolipid metabolism. Therefore, our study provides the basis for the compatibility and efficacy of ASE and GEB.

Triterpenoid Saponins From the Fruit of Acanthopanax senticosus (Rupr. & Maxim.) Harms.

Project description:Five new oleanane-type triterpenoid saponins (1-5), together with 24 known saponins (6-29) were isolated from the fruit of Acanthopanax senticosus. Their structures were determined by extensive spectroscopic analysis, including 1D, 2D nuclear magnetic resonance (NMR), and high-resolution electrospray ionization mass spectrometry (HR-ESI-MS), in combination with chemical methods (acid hydrolysis). The neuroinflammation model was established by lipopolysaccharide (LPS)-induced BV2 microglia, and the neuroprotective effects of all compounds (1-29) were evaluated.

Changes in Growth Performance, Physiological Parameters, and Lipid Metabolism-related Genes Expression of Hybrid Yellow Catfish (Tachysurus fulvidraco ♀ × Pseudobagrus vachellii ♂) following different dietary Acanthopanax Senticosus levels

Project description:High-density aquaculture and over-nutrition may cause fatty liver disease in hybrid yellow catfish, reduce the quality of fish products, and limit the development of the industry. In this experiment, Acanthopanax senticosus powder was used as an additive for hybrid yellow catfish feed. A control group (fed on a diet without A.senticosus) and five groups fed on diets supplemented with A.senticosus (0.5, 1, 2, 4 and 8 g A.senticosus / kg). To study the effects of A.senticosus on the growth performance and physiological parameters of hybrid yellow catfish, and to determine the effect of A.senticosus on the expression of genes related to lipid metabolism in the liver by transcriptome analysis. It was found that dietary supplementation with A.senticosus at 2-4g/kg can promote the growth of yellow catfish, reduce the levels of total cholesterol and triacylglycerol in serum, and increase the activity of fatty acid synthase involved in lipid transport in the liver. Gene expression profiles in the liver were compared between the control group and 4g/kg A.senticosus group, and 5 differentially expressed genes in these groups were identified. Annotation analyses using tools at the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes databases showed that these five differentially expressed genes were mainly involved in the regulation of fat metabolism. The experimental results show that adding a suitable amount of A.senticosus in the diet can reduce the expression of FADS2, ELOVL2, and PLPP3 genes in the liver and stimulate the expression of CYP24a gene, which will reduce the contents of TG and TC in the body, reduce the deposition of fat in the tissue, and promote lipid Metabolism to achieve the purpose of protecting the liver. Therefore, A.senticosus can be used as a healthy feed additive for hybrid yellow catfish, and the appropriate content in the diet is 2-4g/kg.

Gastrodin Derivatives from Gastrodia elata.

Project description:Nine new gastrodin derivatives, including seven p-hydroxybenzyl-modified gastrodin ethers (1-7), 6'-O-acetylgastrodin (8), and 4-[α-D-glucopyranosyl-(1 →6)-β-D-glucopyranosyloxy]benzyl alcohol (9), together with seven known derivatives, were isolated from an aqueous extract of Gastrodia elata ("tian ma") rhizomes. Their structures were determined by spectroscopic and chemical methods as well as single crystal X-ray diffraction. Compounds 1-4, 7, 10, and 11 were also isolated from a reaction mixture by refluxing gastrodin and p-hydroxybenzyl alcohol in H2O. As both gastrodin and p-hydroxybenzyl alcohol exist in the plant, the reaction results provide evidence for the production and increase/decrease of potential effective/toxic components when "tian ma" is decocted solely or together with ingredients in Chinese traditional medicine formulations, though the isolates were inactive in the preliminarily cell-based assays at concentrations of 10 μM. Moreover, using ultra-performance liquid chromatography high-resolution electrospray ionization mass spectrometry (UPLC-HRESIMS), 4, 7, 10, and 11, as well as component variations, were detectable in the freshly prepared extracts of different types of samples, including the freeze-dried fresh G. elata rhizomes.
