Project description:Acute pancreatitis (AP) is an inflammatory disease of pancreatic tissue caused by pancreatic acinar cell injury. However，its miRNA expression is not clear.A mouse model of acute pancreatitis was established by the establishment of Caerulein and LPS, high throughput miRNA sequencing was performed on the pancreatic tissues of mice in the normal group and model group. We found 25 miRNAs with relatively significant differential expression by analyzing the miRNA expression profile. Then we performed downstream target gene prediction for differentially expressed miRNAs. Analysising of KEGG, GO, GSEA and network interaction, it was found that its therapeutic effect was closely related to apoptosis, inflammatory response and autophagy process.

Project description:In the early stages of acute pancreatitis, diagnostic sensitivity is limited and prognosis is difficult to determine. We provide a new idea for the diagnosis and prognosis of AP by studying the expression of plasma miRNAs in patients. Plasma samples from 22 normal subjects and AP patients at different stages were screened for miRNAs sequencing. A total of 27 miRNAs were found to be differentially expressed. Then we performed downstream target gene prediction for differentially expressed miRNAs. Analysising of KEGG, GO, GSEA and network interaction, it was found that its therapeutic effect was closely related to apoptosis, inflammatory response and autophagy process.

Pancreatic cancer and pancreatitis miRNA profiles

Project description:This project analyzes pancreatic tissue profiles of pancreatic cancer patients, pancreatitis patients, and controls. Since miRNAs are known to be valuable diagnostic markers, we asked whether respective patterns of pancreatic cancer patients can be detected in biopsies. The project aimed at an impoved understanding of complex profiles rather than single markers. Thus, a high-throughput technique was necessary, profiling all known miRNAs integratively. Three markers have been validated by using qPCR. n = 22 normal controls, n = 27 pancreatitis samples, and n = 136 pancreatic cancer samples have been screened for the complete miRNA repertoire. Please note that each miRNA has been measured in seven replicates and the median of the replica has been computed.

Transcriptome sequencing and ceRNA (lncRNA-miRNA-mRNA) network construction in cardiac fibrosis and heart failure

Project description:Background: Cardiac fibrosis (CF) and heart failure (HF) are familiar heart diseases that are harmful to health, and severe CF can lead to HF. In this study, we tried to find their common potential molecular markers, which may help the diagnosis and treatment of CF and HF. Methods: Firstly, RNA library construction and high-throughput sequencing were performed. In addition, the DESeq2 package in R was used to screen genes with differentially expressed between different samples. Then, take the intersection of the differential mRNA, miRNA and lncRNA obtained for the two diseases. Thirdly, the ConsensusPathDB (CPDB) was used to perform biological functions enrichment for intersection differentially expressed mRNAs (DEmRNAs). Fourthly, construct gene interaction network and screen out key genes. Fifthly, RT-PCR verification was performed. Lastly, GSE104150 and GSE21125 data sets were utilized to validate the expression and diagnostic analysis. Results: There are 1477 DEmRNAs, 502 differentially expressed lncRNA (DElncRNAs) and 36 differentially expressed miRNA (DEmiRNAs) between CF and healthy control group. There are 607 DEmRNAs, 379 DElncRNAs and 42 DEmiRNAs between HF and healthy control group. CH and FH shared 146 DEmRNAs, 80 DElncRNAs, and 6 DEmiRNAs. Hsa-miR-144-3p, CCNE2, C9orf72, MAP3K20-AS1, LEF1-AS1, AC243772.2, FLJ46284 and AC239798.2 were keys genes in lncRNA-miRNA-mRNA network. In addition, hsa-miR-144-3p and CCNE2 may be considered as potential diagnostic gene biomarkers in CF and HF.
