Transcriptome profile form endometrial biopsy samples of pregnant and non-pregnant cows

Project description:Purpose: characterize the uterine transcriptome profiles of pregnant (P) versus non-pregnant (NP) cows during early pregnancy and attempted to define a potential set of marker genes that can be valuable for predicting pregnancy outcome. Methods: beef cows were synchronized and artificially inseminated at detected estrus. Six days after AI, jugular blood samples and a biopsy from the uterine horn contralateral to the ovary containing the corpus luteum were collected. Based on pregnancy outcome on day 30, samples were retrospectively allocated to the following groups: Pregnant and Non-Pregnant. Both groups had similar plasma progesterone concentrations on D6. Uterine biopsies were submitted to RNA-Seq analysis in a Illumina HiScanSQ platform. Results: The 272,685,768 million filtered reads were mapped to the Bos taurus reference genome and 14,654 genes were analyzed for differential expression between groups. Transcriptome data showed that 216 genes are differently expressed when comparing NP versus P uterine tissue (Padj≤0.1). More specifically, 36 genes were up-regulated in P cows and 180 are up-regulated in NP cows. Conclusions: this study characterized a unique set of genes, expressed in the uterus on 6 days after insemination, that indicate a receptive state leading to pregnancy success. Whole-genome transcriptome profiling in PFOS-treated uterine artery endothelial cells isolated from pregnant women

Project description:During pregnancy, perfluorooctane sulfonate (PFOS) exposure is linked to increased risks of preeclampsia and fetal developmental complications. Although experimental and circumstantial data suggest that PFOS induces endothelial dysfunction, leading to decreased uterine arterial blood flow and gestational hypertension, the precise regulatory mechanisms responsible for this effect remain unknown. To address this issue, we treated human uterine artery endothelial cells (hUAECs) isolated from pregnant women with PFOS and conducted a comparative transcriptomic analyses to understand the underlying mechanism of PFOS-induced endothelial dysfunction.

Effect of depletion of TCF23 on myometrium transcriptome profile during late pregnancy

Project description:TCF23 is a basic-helix-loop-helix transcription factor lacking a DNA binding domain with a C-terminal mediated inhibitory action. Tcf23 was significantly induced by progesterone but not estradiol in vitro and in vivo. To evaluate the biological role of Tcf23, we generated the Tcf23 knockout mouse. Although pregnant Tcf23 KO mice were able to maintain gestation until full term, a considerable proportion of KO females experienced delayed parturition, dystocia, and reduced litter outcomes, with a high incidence of resorption and fetal lethality. Morphological analysis of the pregnant uterus revealed aberrant disruption of both circular and longitudinal myometrial layers and disarrayed extracellular matrix at the conceptus sites in the KO mice. Transcriptome profile analysis of the KO myometrium demonstrated disruptions in cell adhesion, extracellular matrix (ECM), and gap junction signaling, indicating the crucial role of TCF23 in myometrial remodeling and preparation for contractility during late pregnancy. Altogether, our research highlights a novel cause for labor dysfunction mediated by loss of Tcf23 activity in murine uterine tissue.

An epigenetic timer of gestation length in mice [scRNA-Seq]

Project description:Timing mechanisms in biology remain poorly understood. As one prime example, little is known about the mechanisms that specify how long the gestating uterus will remain quiescent before entering labor1. Our lack of insight into this fundamental question, which applies to all mammalian species, also limits investigation into potential causes of preterm labor, a major human pregnancy complication. Here, we provide evidence that gestation length in mice is determined by an epigenetic timer that runs autonomously within the fibroblasts of the pregnant uterus. The timer is set during the peri-implantation period when select genomic loci establish appropriate levels of the repressive histone mark H3K27me3. These loci then progressively lose H3K27me3, thereby scheduling the uterine cell state transitions and associated gene expression changes of late gestation that are the proximal mediators of labor onset. Overwinding of the timer’s peri-implantation setpoint via genetic ablation of the histone demethylase KDM6B delays these transitions and extends gestation length. These results provide insight into how complex tissues employ epigenetic pathways to specify the timing of discrete biological events. We also anticipate that further dissection of the ways such uterine programming controls gestation length may suggest novel approaches for improving human pregnancy outcomes.
