Project description:Isthmin 1 (ISM1) is a secreted 60 kDa protein that exhibits pro-apoptotic and anti-angiogenic functions in experimental settings. We generated ISM1 knockout mice (ISM1-/- ) in C57BL/6J background through CRISPR gene editing technology to understand the physiological function of endogenous ISM1. The ISM1-/- mice presented spontaneous pulmonary emphysema and immune infiltration under the ambient environment. These findings suggest that ISM1 might be an anti-inflammatory protein important for maintaining lung homeostasis. To investigate the role of ISM1 in the lung under steady state, we compare the gene expression in the ISM1 knockout lungs with that of the wild type. Lungs of young adult mice (2 months old) from wild type and ISM1-/- mice were harvested and bulk RNASeq was performed.

Project description:Isthmin 1 (ISM1) is a secreted protein of 60 kDa which has known functions of anti-apoptosis and anti-angiogenesis. To understand the physiological functions of ISM1, we generated Ism1 knockout (Ism1-/-) mice in C57BL/6J background through CRISPR gene editing technology. Under normal conditions, Ism1-/- mice presented spontaneous and progressive lung emphysema with enhanced immune cell infiltration compared to wildtype mice suggesting the role of ISM1 in pulmonary inflammation. In order to understand the pathways perturbed in the Ism1-/- mice, bulk RNA-sequencing analysis was performed using total RNA isolated from the lungs of WT and Ism1-/- mice at post-natal day 7 (P7) stage, when the process of alveolarization occurs.

Project description:To explore potential impact of Aldh1a1 deficiency in the retinoic acid signaling in the lung, gene expression levels were compared between wildtype and Aldh1a1 deficient lung tissues. The expression levels of retinoic acid responsive genes were very similar between wildtype and Aldh1a1 deficient lungs, suggesting that Aldh1a1 deficiency has minimal impact on retinoic acid signaling.
