Project description:High sugar character is one of the important assessment indexes for selecting and breeding excellent sugarcane varieties. In order to screen important proteins that may be involved in sugar synthesis and accumulation during the ripening process of different sugarcane varieties, and to provide theoretical basis for future breeding of sugarcane varieties with high sugar maturity, the early-maturing sugarcane variety GL05-136 and late-maturing sugarcane variety GR2 were used as experimental materials. iTRAQ technique was used to identify the intraspecific and interspecific proteomic differences between the two sugarcane varieties at different sampling periods.

Each dual perturbation experiment consists of four RNA-seq samples (WT, WT+nutrient supplementation, transcription factor KO, transcription factor KO+nutrient supplementation). Project description:Previous reports suggest that the maturation rate of malaria parasites within red blood cells (RBC) is not constant for a given species in vivo. For instance, maturation can be influenced by host nutrient status or circadian rhythm. Here we observed in mice that systemic host inflammation, induced by lipopolysaccharide (LPS) conditioning or ongoing acute malaria infection, slowed the progression of a single cohort of parasites from one generation of RBC to the next. LPS-conditioning and acute infection both triggered substantial changes to the metabolomic composition of plasma in which parasites circulated. This altered plasma directly slowed parasite maturation in a manner that could not be rescued by supplementation, consistent with the presence of inhibitory factors. Single-cell transcriptomic assessment of mixed parasite populations, exposed to a short period of systemic host inflammation in vivo, revealed specific impairment in the transcriptional activity and translational capacity of trophozoites compared to rings or schizonts. Thus, we provide in vivo evidence of transcriptomic and phenotypic plasticity of asexual blood-stage Plasmodium parasites when exposed to systemic host inflammation

Project description:Progression of non-alcoholic fatty liver disease (NAFLD) to non-alcoholic steatohepatitis (NASH) is a major cause of end-stage liver diseases. There is a high need for predictive human ex vivo models to translate preclinical findings for potential new NAFLD drugs to human disease. About a decade ago, precision cut liver slices (PCLSs) have been established as an ex vivo assay for humans and other organisms. The molecular and functional baseline characteristics of PCLSs have been extensively described previously. In the present study, we provide a comprehensive RNA-Seq based molecular characterization of a new assay for NAFLD that induces steatosis in human and mouse PCLSs culture by incremental supplementation of sugars (glucose and fructose), insulin, and fatty acids (palmitate, oleate). Independent of the triglyceride levels of the human donor liver organs at baseline, the nutrient over-supplementation leads to an increase of triglyceride synthesis and cytokine release to the culture supernatant and thus recapitulates hallmark features of steatosis and inflammation in NAFLD. The mirrored design of human vs. mouse liver slices cultured at the exact same conditions allows a comprehensive assessment of donor variability and the contribution of time vs. nutrient factors to the overall observed molecular pattern and the investigation of conserved and species-specific transcriptional pathways that are involved in steatosis and inflammation of the liver.
