Project description:In addition to a series of in vitro experiments characterizing the role of Coxsackie-Adenovirus Receptor (CAR) in erythropoiesis (GSE147963), in vivo proof of concecpt was obtained from an Mx1-Cre Cxadrfl/fl conditional knockout mouse model. After poly I:C injection (400µg on 3 consecutive days) to induce Cxadr deletion mice were observed for up to 10 months to study changes in erythropoiesis. After development of anemia, mice were sacrificed and myeloid BM progenitors sorted. RNA of purified populations was used for RNA-seq to study molecular effector mechanisms.

Proteome and phosphoproteome analysis of autophagy deficient ATG5flox:CamKII-Cre and ATG5flox:VGAT-Cre conditional knockout mice

Project description:The experiment was performed to find selective targets of autophagy in different subtypes of neurons. Therefore, brain tissue of conditional knockout mice lacking autophagy protein ATG5 in inhibitory or excitatory neurons was used. Cortices (ATG5flox:CamKII-Cre) / striata (ATG5flox:VGAT-Cre) of mice were mixed with lys6 labeled brain lysates as internal controls between the samples. Samples were measured by LC-MS/MS and resulting H/L ratios were used to quantify proteomic and phosphoproteomic changes.

Mx1-cre mediated Rgs12 conditional knockout mice exhibit increased bone mass phenotype.

Project description:Regulators of G-protein Signaling (Rgs) proteins are the members of a multigene family of GTPase-accelerating proteins (GAP) for the Galpha subunit of heterotrimeric G-proteins. Rgs proteins play critical roles in the regulation of G protein couple receptor (GPCR) signaling in normal physiology and human diseases such as cancer, heart diseases, and inflammation. Rgs12 is the largest protein of the Rgs protein family. Some in vitro studies have demonstrated that Rgs12 plays a critical role in regulating cell differentiation and migration; however its function and mechanism in vivo is largely unknown. Here, we generated a floxed Rgs12 allele (Rgs12(flox/flox) ) in which the exon 2, containing both PDZ and PTB_PID domains of Rgs12, was flanked with two loxp sites. By using the inducible Mx1-cre and Poly I:C system to specifically delete Rgs12 at postnatal 10 days in interferon-responsive cells including monocyte and macrophage cells, we found that Rgs12 mutant mice had growth retardation with the phenotype of increased bone mass. We further found that deletion of Rgs12 reduced osteoclast numbers and had no significant effect on osteoblast formation. Thus, Rgs12(flox/flox) conditional mice provide a valuable tool for in vivo analysis of Rgs12 function and mechanism through time- and cell-specific deletion of Rgs12.

Expression data from Dmp1-Cre targeted and non-targeted Cxcl12-abundant reticular (CAR) cells in mouse bone marrow

Project description:In mouse bone marrow, mesenchymal stem cells (MSC) has the potential to form osteocytes, adipocytes and cartilage. In the process of osteogenesis, MSCs differenetiate into stromal cells, such as CAR cells. Osteoblast is responsible for the formation of osteocytes and osteoblasts may be differentiated from a subset of CAR cells. Dmp1-Cre targeted CAR cells are thought to enrich for a osteoblast progenitor population. We used microarrays to detail the gene expression profiles among Dmp1-Cre targeted and non-targeted CAR cells. Gene expression diffferences were compared to support the hypothesis that Dmp1-Cre targeted CAR cells may be enriched for osteoblast progenitors. Dmp1-Cre targeted and non-targeted CAR cells were FACS sorted from three mice. RNA were extracted from these sorted cells and processed for microarray using Affymetrix mogene 1.0 ST chip.
