We have shown PRMT1 is a protein that is upreguated in and promotes vaious cancers. The expression of its gene is localized to embryonic neural cells during vertebrate embryogenesis. The project is to identify the interaction partners of PRMT1, by which PRMT1 regulates neural stemness in both cancer cells and neural stem cells.

Comparison study of glioma stem cells and adult human neural stem cells

Project description:Glioblastoma, the most common and malignant brain tumor, harbors stem-like cells that self-renew and propagate upon serial transplantation. Although they share functional, morphological and developmental similarities to adult brain neural stem cells, stem cell characteristic pathways contributing to glioblastoma stem-like cells have not been consistently determined. Towards this goal we have provided an internally coherent molecular reference that compares adult neural and glioblastoma stem cells cultivated under identical conditions. Genes dysregulated between these populations are correlated with clinical outcome in glioblastoma, and are highly expressed in embryonic and induced pluripotent stem cells. The resource yields a key role for Wnt and a core group of dysregulated pathways. We have verified the contribution to proliferation and sphere formation of Wnt- differentially activated genes through the Wnt inhibitor SFRP1. The glioblastoma and neural stem cell gene-expression and pathway comparative resource provides a powerful new tool for the identification of potential therapeutic targets. 14 samples was analyzed; 5 individual samples of adult neural stem cells and 9 individual samples of glioma stem cells

Project description:The orphan nuclear receptor Nurr1 has been shown to be critical for the development of ventral midbrain dopaminergic neurons. Consequently, the development of ES cells overexpressing Nurr1 has raised hope for the development of cell replacement therapies for Parkinson's Disease to replace degenerated dopaminergic neurons. However, the molecular consequences of Nurr1 on gene expression in these cells remain unknown. To address this, stable, clonal, c17.2 neural stem cell lines were established that overexpressed the orphan nuclear receptor Nurr1 (clone 42 & clone 48) or parental control cell line (puroB & puroD, respectively). Modification of Disp3 expression levels by CRISPR/Cas9 technology affects neural stem cells capacity to differentiate.

Project description:DISP3 (PTCHD2), a sterol-sensing domain-containing protein, is highly expressed in neural tissue but its role in neural differentiation is unknown. To determine the effect of DISP3 on neural stem cells phenotype of NS-5 cells, we impaired Disp3 gene using CRISPR/Cas9 technology. Modification of Disp3 expression levels affects NS-5 capacity to differentiate. In differentiated Disp3 sgRNAs cell cultures we observed higher number of highly differentiated astrocytes and oligodendrocytes and neurons with not only increased Î²III-tubulin mRNA expression but also with more developed neurites.

The protein profiles of neural stem cells

Project description:Stemazole was reported to induce the survival of human neural stem cells in the absence of growth factors and to have therapeutic effects on neurodegenerative diseases. However, the molecular mechanisms of stemazole against apoptosis are ambiguous. In this study, tandem mass tag (TMT)-based proteomics was performed to obtain whole protein expression profiles of human neural stem cells in different groups, which was performed with or without stemazole and under extreme conditions. Bioinformatics analysis based on protein–protein interaction (PPI) network construction, gene ontology (GO) and Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathway enrichment analysis were adopted to explore crucial proteins and possible pharmaco-logical mechanisms. A total of 77 differentially expressed proteins were identified, comprising 38 upregulated proteins and 39 downregulated proteins.
