Project description:SigE is a sigma factor found in Streptomyces species and is the key regulator of cell envelope stress response in Streptomyces coelicolor. To determine the regulon of SigE under conditions of cell envelope stress, 10 ug / ml vancomycin was used to induce cell envelope stress in the wild type and a sigE deletion strain of S. coelicolor. Samples were taken 0, 30, 60 and 90 minutes after addition of vancomycin and subjected to transcriptional profiling using Affymetrix arrays. Note – equivalent data for wild type Streptomyces coelicolor M600 is submitted as part of E-MEXP-3032. The data for both M600 (E-MEXP-3032) and J2130 (this submission) were generated at the same time employing the same protocols.

Response of Streptomyces coelicolor to treatment with the antibiotics vancomycin bacitracin and moenomycin A

Project description:To determine the response at the transcriptional level of S. coelicolor cells to treatment with 3 antibiotics that target distinct stages of cell wall biosynthesis, biological triplicate cultures of strain M600 were treated with sub-lethal concentrations (10 µg/ml) of vancomycin, bacitracin or moenomycin A. Samples were taken 0, 30, 60, 90 minutes after addition of the antibiotic. A negative control which received no drug treatment was also performed.

Transcription profiling of Streptomyces ceolicolor M600 and J2139 (sigR deletion) strains

Project description:In Streptomyces coelicolor the SigR sigma factor controls genes involved in coping with the deleterious formation of disulphide bonds in cellular proteins. This experiment provides global gene expression patterns of M600 and J2139 (sigR deletion) grown in NMMP plus glucose medium to mid-log phase then treated with diamide. The data reveal that sigR activates, directly or indirectly, more than 65 transcription units, encoding functions that include thiol-disulphide oxidoreductases, thiol buffers, and ATP-dependent proteases. The sigR regulon partially overlaps the ClgR and HspR regulons, that include functions for protein degradation and protein refolding. Diamide also leads to the transient down-regulation of central metabolic gene expression, in particular ribosomal protein expression.

Identification by ChIP-Seq of the binding sites of the extracytoplasmic function (ECF) sigma factor SigE in the genome of Streptomyces coelicolor A3(2) M600.

Project description:SigE is a sigma factor found in Streptomyces species and is the key regulator of cell envelope stress response in Streptomyces coelicolor. This ChIP-Seq experiment was carried out to determine the binding sites of SigE under conditions of cell envelope stress induced by 10 ug / ml vancomycin for 30 minutes. A 3xFLAG tagged SigE was introduced in a SigE deletion strain and ChIP-Seq was carried out using M2 (Sigma Aldrich A2220) gel suspension. Non-immunoprecipitated (total) DNA preparations were used for background determination and the wild type M600 was used as the negative control.
