Project description:Since Anopheles gambiae female mosquitoes mate only once in their lifetime, and store sperm received from males potentially for weeks, genes and pathways regulated by mating in the sperm storage organ (spermatheca) to preserve sperm function may represent new targets for vector control. The transcriptional response to mating in the spermatheca was investigated using Agilent two-color microarrays. Spermathecae from mated females were collected 24 h after mating and compared to age-matched virgins. 4 biological replicates were performed.

Transcription profiling by array of M and S molecular forms of Anopheles gambiae at each of three developmental stages

Project description:We examined patterns of gene expression in two independent colonies of both M and S molecular forms of Anopheles gambiae at each of three developmental stages of interest: late larvae, sugar-fed virgin females, and gravid females. For each colony, replicates were derived from independent RNA samples extracted from different cohorts to ensure that trends were reproducible. In addition, each replicate was derived from larvae (adults) drawn from three pans (cages) to minimize the contribution of any individual pan to variation between samples. Data were obtained from a total of five biological replicates per mosquito colony.

Project description:To gain better insight into the molecular link between the post-mating transcriptional and morphological changes and the behavioral and physiological shift induced by copulation in An. gambiae females, we performed an in-depth transcriptional analysis of the female lower reproductive tract (LRT), comprising the atrium and the spermatheca, to identify the molecular pathways specifically regulated in these female reproductive tissues. We analyzed three time points post mating (3, 12 and 24 hours post mating) in order to capture transcriptional changes occurring within a broad time window after copulation. For each time point, tissues were collected from mated and age-matched virgin females and the transcriptional profile compared across 4 biological replicates using Agilent one-color microarrays.

Project description:We performed genome-wide profiling experiments on tissues from Drosophila melanogaster adult females with a mutation in the p24 gene logjam (loj) to test the hypothesis that loss of loj function causes a transcriptional response characteristic of ER stress activation. Experiment Overall Design: We compared expression profiles from loj mutant females to those of controls. We compared mutant and control abdomen tissue as well as mutant and control tissue preparations from the remainder of the fly (the head/thorax). Three mutant and three control RNA samples for each set of tissue were used for array hybridization (12 total array samples).

Organ-specific gene expression changes in the fetal liver and placenta in response to maternal folate depletion

Project description:Growing evidence supports the hypothesis that the in utero environment can have profound implications for fetal development and for offspring health in later life. Current theory suggests that conditions experienced in utero prepare, or ‘programme’, the fetus for its anticipated post-natal environment. The mechanisms responsible for these programming events are poorly understood but are likely to involve gene expression changes. Folate is essential for normal fetal development and inadequate maternal folate supply during pregnancy has long term adverse effects on the offspring. We tested the hypothesis that inadequate folate supply during pregnancy alters offspring programming through altered gene expression. Female C57BL/6J mice were fed diets containing 2 mg folic acid/kg or 0.4 mg folic acid/kg for 4 weeks before mating and throughout pregnancy. At 17.5 day gestation, genome-wide gene expression in fetal liver and placenta of male offspring was measured by microarray analysis. In the fetal liver, 989 genes (555 up-regulated, 434 down-regulated) were expressed differentially in response to maternal folate depletion, with 460 genes expressed differentially (250 up-regulated, 255 down-regulated) in the placenta. Only 25 differentially expressed genes were common between organs, revealing that maternal folate intake during pregnancy influences fetal gene expression in a highly organ specific manner which, we propose, reflects prioritised protection of essential organ-specific functions.
