Project description:Changes in gene expression in P. trichocarpa leaves was measured in response to growth competition by P. x canescens.Square petri dishes (12 x 12 cm) were filled autoclaved sand, 3 plantlets and Woody Plant Medium (WPM). In each petri dish we are growing 3 plantlets. As control 3 plantlets of P. trichocarpa and for the experiment a mixture of P. canescens and P. trichocarpa were P. trichocarpa is all the time the middle plantlet.

Abscisic acid (ABA) application to Populus × canescens in hydroponics

Project description:4 weeks old rooted plantlets of P. × canescens (Clone INRA717 1-B4) were cultivated in hydroponics in 2 l pots in Long Ashton nutrient solution in a culture room for 8 weeks before treatments started. Three treatments were applied to the plants: control treatment (-ABA), continuous 100 µM ABA treatment (+ABA) and discontinuous 100 µM ABA treatment (±ABA). ABA was fed to +ABA plants during the whole treatment period of 30 days. ABA was fed to ±ABA plants for three days in two weeks. Developing xylem and mature xylem were collected separately during the harvest and shortly frozen in liquid nitrogen. RNA was extracted from these samples and followed by RNA-sequencing.

Project description:Arabidopsis plants were grown in sand and supplied with full nutrient during 6 to 7 weeks in a growth chamber in controlled conditions (21/18°C day/night temperature, 60 % humidity, 8 h photoperiod). Then for the split-root experiments, the root system was washed in water to discard the sand and separated in two parts dipped in 2 containers (300 mL each) containing either the same medium (MS/10 supplied with 500 ?M Pi (cP500) or without Pi (cP0) or different media (500 ?M Pi and no Pi, respectively spP500 and spP0) for two days. Three plants were pooled for each condition and the experiment was triplicated.

Transcription profiling of pistils from Arabidopsis wild type, ant-4 mutant and ino-1mutant plants at different developmental stages

Project description:This experiment was designed to identify genes expressed preferentially in the two integuments of the Arabidopsis ovule. Pistils from wild type and two ovule mutants were compared against each aintegumenta-4 (ant-4) which lacks both integuments and inner no outer (ino-1) which lacks the outer integument. Genes that are highly expressed only in the integuments were expected to be reduced in expression in the mutants, as compared with wild type. Pistils containing ovules through all stages of ovule development prior to pollination were pooled for one experiment (FULL arrays), and for two separate experiments, a set of early differentiation stages (EARLY arrays) and a set of later differentiation stages (LATE ARRAYS) were pooled. Wild type and mutant lines are in the ecotype Landsberg erecta.
