Krüppel-like factor 4 is a transcription factor with anti-proliferative effects in differentiated cells, but with the ability to reprogram adult cells into cell-cycling pluripotent cells. Project description:To analyse roles of HAI-1/Spint1 in intestinal tumorigenesis, we examined the effect of intestine-specific deletion of Spint1 gene on Apc(Min/+) mice. The loss of Hai-1/Spint1 significantly accelerated tumor formation in ApcMin/+ mice and shortened their survival periods. Mouse small intestine tumor tissue or background mucosa lacking macroscopically visible tumors were proceeded to RNA extraction and hybridization on microarrays (Affymetrix Mouse Genome 430 2.0 Array). Non-tumor or tumor intestinal mucosa tissues of Apc (Min/+)/Spint1 (flox/flox) mice and non-tumor or tumor intestinal mucosa tissues of Apc (Min/+)/Spint1 (flox/flox)/Vil-Cre mice were analysed. The experiment was repeated respectively.

Colony stimulating factor (CSF1) treatment of kidneys that have undergone ischemia reperfusion injury

Project description:Mice were subjected to renal artery clamps to induce ischemia/reperfusion injury to the kidney. 3 days post surgery mice were injected with CSF1 or PBS daily for a further 3 days. Mice were sacrificed 5 and 7 days post surgey and the kidney was removed and homogenised into single cell suspensions. Cells were selected on their expression of CD45, CD11b markers and the lack of CD11c. These cells were then subjected to microarray analyses to determine the global gene expresssion profiles of mice with and without treatment.

Project description:Plant cell wall degrading enzymes (PCWDEs) are key virulence determinants in the pathogenesis of the potato pathogen, Pectobacterium atrosepticum. In this study, we report the impact on virulence of a transposon insertion mutation in the metJ gene that codes for the repressor of the methionine biosynthesis regulon. In a mutant strain defective for the small regulatory RNA rsmB, PCWDEs are not produced and virulence in potato tubers is almost totally abolished. However, when the metJ gene is disrupted in this background, the rsmB- phenotype is suppressed and virulence and PCWDE production are restored. Additionally, when metJ is disrupted, production of the quorum sensing signal, N-(3-oxohexanoyl)-homoserine lactone, is induced precociously. The metJ mutant strains showed pleiotropic transcriptional impacts affecting approximately a quarter of the genome. Genes involved in methionine biosynthesis were most highly up-regulated, but many virulence-associated transcripts were also upregulated. This is the first report of the impact of the MetJ repressor on virulence in bacteria.
