Transcription profiling of degreening transgenic Arabidopsis over time

Project description:Transgenic Arabidopsis thaliana bearing a synthetic degreening circuit were induced to degrade their chlorophyll, and the transcriptomes were profiled at 0, 24, and 48h after degreening induction, as well as in 48h plants that were subsequently allowed to "regreen". Induction of transgene expression resulted in overexpression of the chlorophyll degrading enzymes chorophyllase and PAO (pheide a oxygenase), as well as expression of a diRNA construct to the chlorophyll biosynthetic POR enzymes (NADPH:protochlorophyllide oxidoreductase).

Project description:Three 2cm segments were excised from different parts (TOP, MID, BOT) along the vertical axis of a 4 week old (25cm) stem of flax (L. usitatissimum) were compared using a cDNA amplicon array. Each segment represented a different developmental stage, especially in relation to bast fibre differentiation (i.e. TOP= elongation, MID=transition, BOT= thickening). Only the cDNAs that showed the highest differential expression were sequenced.

Project description:We were interested in determining what genes might be controlled by TFAP2C and/or TFAP2A, either directly or indirectly through regulation of ER-alpha and potentially other signaling pathways. We performed an microarray analysis in MCF7 cells with elimination of either TFAP2C or TFAP2A. The patterns of gene expression with alteration of TFAP2 activity were compared to changes in expression induced by estrogen exposure. Knock-down of TFAP2C in the presence of estrogen altered the pattern of several known ERalpha-regulated genes and a number of genes outside the estrogen-regulated pathways. microRNA profiling by array of human skin melanoma and microenvironment

So they are involved in many cellular processes.
