Ligand-controlled cobalt-catalyzed regiodivergent hydroboration of aryl,alkyl-disubstituted internal allenes.

Project description:We report a stereoselective regiodivergent hydroboration of aryl,alkyl-disubstituted internal allenes with pinacolborane (HBpin) in the presence of cobalt catalysts generated from bench-stable Co(acac)2 and bisphosphine ligands. An interesting correlation between the regioselectivity of this hydroboration and the bite angles of bisphosphine ligands was identified. When hydroboration was conducted with cobalt catalysts containing bisphosphines with medium bite angles (e.g. 98° for dppb and 96° for dppf), HBpin was selectively added to the alkyl-substituted double bond. However, HBpin was selectively added to the aryl-substituted double bond when the reactions were conducted with cobalt catalysts containing bisphosphines with large bite angles (e.g. 111° for xantphos and 114° for Nixantphos). A range of internal allenes underwent these Co-catalyzed hydroboration reactions in a regiodivergent manner to yield the corresponding (Z)-alkenylboronates in high isolated yields and with high regioselectivity. These reactions show good functional group compatibility and can be readily scaled up to gram scales without using a dry box. In addition, the comparison of regioselectivity between the Co-catalyzed hydrosilylation and hydroboration reactions of the same allene substrate suggests that this Co-catalyzed regiodivergent hydroboration of allenes proceeds through a Co-Bpin intermediate. Deuterium-labeling experiments suggest that the Co-Bpin intermediates react with allenes to form allylcobalt species which then react with HBpin to release (Z)-alkenylboronate products.

Ligand-Controlled Regiodivergent Enantioselective Rhodium-Catalyzed Alkene Hydroboration.

Project description:Regiocontrol in the rhodium-catalyzed boration of vinyl arenes is typically dominated by the presence of the conjugated aryl substituent. However, small differences in TADDOL-derived chiral monophosphite ligands can override this effect and direct rhodium-catalyzed hydroboration of β-aryl and β-heteroaryl methylidenes by pinacolborane to selectively produce either chiral primary or tertiary borated products. The regiodivergent behavior is coupled with enantiodivergent addition of the borane. The nature of the TADDOL backbone substituents and that of the phosphite moiety function synergistically to direct the sense and extent of regioselectivity and enantioinduction. Twenty substrates are shown to undergo each reaction mode with regioselectivity values reaching greater than 20:1 and enantiomer ratios reaching up to 98:2. A variety of subsequent transformations illustrate the potential utility of each product.

Ruthenium-Catalyzed trans-Hydroalkynylation and trans-Chloroalkynylation of Internal Alkynes.

Project description:[Cp*RuCl]4 catalyzes the addition of iPr3SiC≡CX (X = H, Cl) across internal alkynes with formation of 1,3-enyne or 1-chloro-1,3-enyne derivatives, respectively; the reaction follows an unorthodox trans-addition mode. The well-balanced affinities of the different reaction partners to the ruthenium catalyst ensure that crossed addition prevails over homodimerization of the individual components, as can be deduced from spectroscopic and crystallographic data of various intermediates; this includes a dinuclear complex in which an internal alkyne bridges two [Cp*RuCl] fragments.

Copper-Catalyzed Regiodivergent Internal Allylic Alkylations.

Project description:We report a copper-catalyzed, regioselective, and stereospecific alkylation of unbiased internal allylic carbonates with functionalized alkyl and aryl Grignard reagents. The reactions exhibit high stereospecificity and regioselectivity for either SN 2 or SN 2' products under two sets of copper-catalyzed conditions, which enables the preparation of a broad range of products with E-alkene selectivity. Density functional theory calculations reveal the origins of the regioselectivity based on the different behaviors of homo- and heterocuprates.

Pd/C-catalyzed regiodivergent hydrocarboxylation and esterification of alkynes.

Project description:An unprecedented and highly reactive Pd/C catalytic system has been introduced for the regiodivergent hydrocarboxylation of terminal alkynes to selectively afford various acrylic and cinnamic acids employing oxalic acid as a CO source as well as a promoter for the formation of the active Pd-H complex. Herein, the formation of cinnamic acid is proposed to follow a unique anti-Markovnikov hydroiodination mechanism and the formation of acrylic acid might follow the traditional hydrocarboxylation pathway. Additionally, internal alkynes undergo hydrocarboxylation and carbonylative esterification with aliphatic alcohols to yield different α,β-unsaturated acids and esters respectively. The designed strategies were successfully leveraged for a diverse class of α,β-unsaturated acids and esters with excellent selectivity and yields under mild reaction conditions. Furthermore, the acid functionalization of complicated naturally derived alkynes, utilizing economical and bench-stable oxalic acid and a commercially accessible reusable catalyst with gram-scale applicability are the additional benefits of the established protocol.

Hydroboration of Terminal Alkenes and trans-1,2-Diboration of Terminal Alkynes Catalyzed by a Manganese(I) Alkyl Complex.

Project description:A MnI -catalyzed hydroboration of terminal alkenes and a 1,2-diboration of terminal alkynes with pinacolborane (HBPin) is described. For alkenes, anti-Markovnikov hydroboration takes place; for alkynes the reaction proceeds with excellent trans-1,2-selectivity. The most active pre-catalyst is bench-stable alkyl bisphosphine MnI complex fac-[Mn(dippe)(CO)3 (CH2 CH2 CH3 )]. The catalytic process is initiated by migratory insertion of a CO ligand into the Mn-alkyl bond to yield an acyl intermediate, which undergoes B-H bond cleavage of HBPin (for alkenes) and rapid C-H bond cleavage (for alkynes), forming the active MnI boryl and acetylide catalysts [Mn(dippe)(CO)2 (BPin)] and [Mn(dippe)(CO)2 (C≡CR)], respectively. A broad variety of aromatic and aliphatic alkenes and alkynes was efficiently and selectively borylated. Mechanistic insights are provided based on experimental data and DFT calculations revealing that an acceptorless reaction is operating involving dihydrogen release.

Copper-Catalyzed Triboration of Terminal Alkynes Using B2 pin2 : Efficient Synthesis of 1,1,2-Triborylalkenes.

Project description:We report herein the catalytic triboration of terminal alkynes with B2 pin2 (bis(pinacolato)diboron) using readily available Cu(OAc)2 and Pn Bu3 . Various 1,1,2-triborylalkenes, a class of compounds that have been demonstrated to be potential matrix metalloproteinase (MMP-2) inhibitors, were obtained directly in moderate to good yields. The process features mild reaction conditions, a broad substrate scope, and good functional group tolerance. This copper-catalyzed reaction can be conducted on a gram scale to produce the corresponding 1,1,2-triborylalkenes in modest yields. The utility of these products was demonstrated by further transformations of the C-B bonds to prepare gem-dihaloborylalkenes (F, Cl, Br), monohaloborylalkenes (Cl, Br), and trans-diaryldiborylalkenes, which serve as important synthons and have previously been challenging to prepare.

Ni-catalyzed hydroaminoalkylation of alkynes with amines.

Project description:Allylic amines are versatile building blocks in organic synthesis and exist in bioactive compounds, but their synthesis via hydroaminoalkylation of alkynes with amines has been a formidable challenge. Here, we report a late transition metal Ni-catalyzed hydroaminoalkylation of alkynes with N-sulfonyl amines, providing a series of allylic amines in up to 94% yield. Double ligands of N-heterocyclic carbene (IPr) and tricyclohexylphosphine (PCy3) effectively promote the reaction.
