ABSTRACT: The title compound, C(24)H(41)NO, is a new derivative of the anti-HIV steroid 17?-(N-tert-butyl-amino-carbon-yl)androst-4-en-3-one. There are four rings in the structure and these are trans-fused. The three six-membered rings exhibit chair conformations, while the five-membered ring adopts an envelope conformation.

Project description:In the title compound, C(24)H(39)NO(2)·H(2)O, the A and C rings of the pregnolene derivative sterol adopt chair conformations, with the B ring in a flattened chair conformation and the five-membered ring in an envelope conformation twisted about the C/D ring junction. The N-tert-butyl-carboxamide substituent is equatorial. The 3?-hydr-oxy H atom and one H atom of the water mol-ecule are disordered over two positions with equal occupancies. In the crystal structure, O-H?O hydrogen bonds between the 3?-hydr-oxy groups of neighbouring mol-ecules form dimers in the bc plane and these dimers are stacked along the a axis by additional O-H?O hydrogen bonds involving the water mol-ecules. The steric effect of the bulky tert-butyl substituent in the carboxamide chain precludes hydrogen-bond formation by the N-H group.

The triiodide ion has near-symmetric linear geometry, with bond lengths of 2.9105?(4)?Å (I-I) and a bond angle of 177.55?(3)° (I-I-I). For this room-temperature structure, the butyl group on the pyridine ring is disordered and has been treated as a rigid rotator, modeled in three separate positions with 1/3, 1/3, 1/3 occupancies. The cations assemble into dimeric forms by way of N-H?N hydrogen bonds.

Metabolically Stable tert-Butyl Replacement.

Project description:Susceptibility to metabolism is a common issue with the tert-butyl group on compounds of medicinal interest. We demonstrate an approach of removing all the fully sp(3) C-Hs from a tert-butyl group: replacing some C-Hs with C-Fs and increasing the s-character of the remaining C-Hs. This approach gave a trifluoromethylcyclopropyl group, which increased metabolic stability. Trifluoromethylcyclopropyl-containing analogues had consistently higher metabolic stability in vitro and in vivo compared to their tert-butyl-containing counterparts.

Project description:A new polymorph of the title compound, [Pd(2)(C(8)H(18)P)(2)(C(8)H(19)P)(2)], has been found. It belongs to the triclinic P-1 space group, whereas the known form [Leoni, Sommovigo, Pasquali, Sabatino & Braga (1992 ?), J. Organo-met. Chem.423, 263-270] crystallizes in the monoclinic C2/c space group. The title compound features a dinuclear palladium complex with a planar central Pd(2)(?-P)(2) core (r.m.s. deviation = 0.003?Å). The Pd-Pd distance of 2.5988?(5)?Å is within the range of a Pd(I)-Pd(I) bond. The mol-ecules of both polymorphs are located on a crystallographic centre of inversion. The mol-ecular conformations of the two polymorphs are essentially identical. The crystal packing patterns, on the other hand, are slightly different.
