Sunitinib: effects of oral administration on haemodynamic and electrocardiographic parameters in the chronically-instrumented conscious guinea-pig using telemetry


ABSTRACT: Conscious guinea-pig chronically-instrumented with a radiotelemetry implant. N=7 Male Dunkin-Hartley guinea-pigs, dosed with Sunitinib at 10, 30, 90 and 180 mg/kg, po, in successive treatments sessions (cross-over design, wash-out = 48h minimum). PK phase: on 9 satellite non-instrumented animals (n=3 animals per group) at T120, T360, T600 and T1440. Haemodynamic parameters (SAP, DAP, MAP, HR). Lead II ECG (RR, PR, QRS, QT, QTc intervals). Arterial blood pressure, heart rate and ECG parameters were measured just before administration (baseline = T0) and 30 (T30), 60 (T60), 90 (T90), 120 (T120), 180 (T180), 240 (T240), 360 (T360), 480 (T480), 720 (T720), 960 (T960), 1200 (T1200) and 1440 min (T1440) post-administration.
