Expression data of murine dermal macrophages

Staphylococcus aureus-induced proteomic changes in the mammary tissue of rats: A TMT study

Project description:Staphylococcus aureus is one of the most important pathogens causing mastitis in dairy cows. The objective of this study was to establish a rat mastitis model induced by a S.aureus infection and to explore proteins that change in proteomes of mammary tissue in different udders states,which provided a better understanding of the host immune response to S. aureus mastitis. Six 72 h postpartum rats were randomly divided into two groups (n=3), with either 100 µL of PBS (blank group) or a S. aureus suspension containing 2×107 CFU·ml−1 (challenge group) infused into the mammary gland duct,respectively. After 24 h of infection, the rats were sacrificed, and mammary gland tissue was collected. A tandem mass tag (TMT)-based technology was applied to compare the proteomes of healthy and mastitic mammary tissue. Compared with the control group, the challenge group had 555 proteins with significant differences in expression, of which 428 were significantly up-regulated (FC>1.2 and p<0.05) and 127 were down-regulated (FC>0.83 and p<0.05 or p<0.01). Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses revealed that up-regulated differentially significant expressed proteins (DSEPs) were associated with mainly immune responses,including integrin alpha M,inter-α-trypsin inhibitor heavy chain 4,alpha-2-macroglobulin.

Project description:The humoral immune response of Galleria mellonella exposed to Mycobacterium bovis BCG (vaccine strain), 1x107 CFU (10 ul inoculum) were injected into the larvae. For low dose BCG challenge, 1x105 CFU (10 ul) were injected. For larval challenge with Actinobacillus pleuropneumoniae (APP), ~1.5x106 CFU (10 ul) were injected. APP were only challenged at this single dose. For larval challenge with Control (0h) were included. APP and low dose (1x105 CFU) BCG were sampled only at 48 h post infection.

The effects of Mkp-1 knockout and systemic E. coli infection on the transcriptome of the liver

Project description:Previously, I have found that Mkp-1 knockout exacerbates inflammation, perturbs lipid metabolism, increases organ damage, mortality, and bacterial burdens. To understand the role of Mkp-1 in the regulation of host defense, we performed RNA-seq analysis using total RNA isolated from livers of uninfected and E. coli-infected wildtype and Mkp-1 knockout mice. Specifically, Mkp-1 wildtype and knockout (both on the C57/129 background) mice were infected with E. coli (O55:B5, ATCC 12014) at a dose of 2.5×107 CFU/g of body weight i.v., or given same volume of PBS. Mice were sacrificed 24 h post-infection to harvest organs. Livers were frozen clamping in liquid nitrogen, and stored at -80°C . Total RNA was isolated with Trizol and used for RNA-seq. RNA-seq results indicate that both E. coli infection and Mkp-1 knockout caused profound changes in the transcriptomes in the mouse livers.

The infection dose was 107 cfu/mouse.

Host response to S. pneumoniae TIGR4 and ΔpotABCD

Project description:The aim of this experiment is to determine the difference in protein expression, if any, in a mouse model of pneumococcal pneumonia using wild type and a polyamine transport operon deletion mutant strain of S. pneumoniae TIGR4, using expression proteomics. Eight week old female C57bl/6 mice were used in this study. The experimental design had two groups of 3 animals each that were administered 50 µL 1 X 107 CFU wild type S. pneumoniae TIGR4 or S. pneumoniae TIGR4 ΔpotABCD strain by intranasal instillation. The control group with three animals received 50 µL PBS. Mice were euthanized 4 hr and 12 hr post infection and lung tissues were harvested. The control group animals were euthanized 12 hr post infection.
