Influence of Dnmt3a knockout on gene expression of preleukemia B cell and CLL cells

ABSTRACT: Although somatic mutations in the DNA methyltransferase 3A (DNMT3A) in chronic lymphocytic leukemia (CLL) are rare, DNMT3A transcript and protein expression is frequently reduced in CLL patients, and previous studies have associated low DNMT3A expression with more aggressive disease and poorer survival.

Project description:Genomic profiles of CLL (Chronic Lymphocytic Leukemia) patients. 11 CLL patients were selected for detection of genomic aberrations, 8 patients with atypical CLL and 3 patients with typical CLL. Patient's DNA were hybridized against Promega control on Agilent G4410A arrays and scanned with the Agilent G2505B scanner.

Project description:Stabilizing mutations of NOTCH1 have been identified in about 10% of chronic lymphocytic leukemia (CLL) cases at diagnosis, with a higher frequency in unmutated IGHV (IGHV-UM) CLL, chemorefractory CLL and CLL in advanced disease phases. Clinically, the presence of NOTCH1 mutations is an independent predictor of overall survival in CLL and associates with resistance to anti-Cd20 immunotherapy. The Gene Expression Profile was generated to identify the peculiar molecular signatures of NOTCH1 mutated CLL in the context of IGHV-UM CLL.
