Single-cell transcriptomic profiles of CD45-negative cells from Mx1Cre-Rbpjf/f mice

ABSTRACT: Induced abltion of Adam10 in Mx1-Cre x Adam10f/f mice results in T cell-independent Inflammatory alopecia. To determine if the Notch signaling pathway was downstream of Adam10, Mx1-Cre x Rbpjf/f mice were generated. After poly(I:C) injection, mouse skin was dissociated and were sorted for CD45-negative cells which were subjected to single-cell RNA sequencing analysis.We found that the ADAM10-Notch signaling axis in type I/III interferon-responsive upper hair follicles (HF) was crucial for the expression of genes that regulated skin microbiota and protected HFs and its stem cell niche from inflammatory destruction.
