Single-cell transcriptomic profiles of CD45-negative cells from WT (C57BL/6), antibiotics non-treated Mx1Cre-Adam10f/f and antibiotics treated Mx1Cre-Adam10f/f mice

ABSTRACT: Induced abltion of Adam10 in Mx1-Cre x Adam10f/f mice results in T cell-independent Inflammatory alopecia. To determine if dysbiosis is a prerequisite for the development of alopecia, Mx1-Cre x Adam10f/f mice were treated orally with an antibiotic cocktail pre- and post-induction. 30 days after poly(I:C) injection, mouse skin was dissociated and were sorted for CD45-negative cells which were subjected to single-cell RNA sequencing analysis.We found that the ADAM10-Notch signaling axis in type I/III interferon-responsive upper hair follicles (HF) was crucial for the expression of genes that regulated skin microbiota and protected HFs and its stem cell niche from inflammatory destruction.
