Genome-wide gene expression analysis of BQ.Ncf1m1J mutated and BQ wild type mice during collagen induced arthritis

ABSTRACT: Whole blood and spleen tissue was collected 15 (d15) or 44 (d44) days postimmunization from mice immunized with type II collagen on day 0 and immunostimulated on day 21. We compared the gene expression profiles of disease prone BQ.Ncf1m1J mice to that in BQ wild type mice. The samples were collected in two sets (marked I and II). The results show a pronounced interferon signature and increase in inflammatory transcripts in the BQ.Ncf1m1J mice.

Project description:Whole blood and spleen tissue was collected from naïve untreated mice. We compared the gene expression profiles of naïve autoimmunity prone BQ.Ncf1.m1J mutated mice and wild type BQ mice. The samples from specific pathogen-free animal facility were collected in two sample sets (marked I and II respectively) and the germ-free facility samples (marked GF / germ free) formed the third analyzed batch. The data revealed a pronounced type I interferon signature in the Ncf1.m1J mutated mice housed in both conditions.

Bulk RNA-seq on paws of collagen-induced arthritis (CIA) mouse model at different disease stages

Project description:The goal of this experiment is to profile longitudinal transcriptome changes during the development of murine arthritis in CIA mouse model. Male DBA1 mice (age between 10 to 12 weeks) were used in this study. CIA was induced by two immunizations with Type II collagen in Complete Freund’s adjuvant (CFA) at day 0, and with Type II collagen in Incomplete Freund’s adjuvant (IFA) at day 21. Bulk RNA-seq were done on right hind paws from control (non-immunized DBA1 mice) and CIA mice at different disease stages (up to 10 weeks after 1st immunization).

Project description:We found that a number of Tfh cells downmodulated BCL6 protein after their development, and we sought to compare the gene expression between BCL6-hi Tfh cells and BCL6-low Tfh cells. CD4+ T cells were sorted from immunized and non-immunized mice for RNA extraction and hybridization on Affymetrix microarrays. Bcl6yfp/+ OT-II cells were transferred to congenic recipient mice, and immunized with NP-OVA in CFA subcutaneously. Seven or ten days after immunization, cells were collected from draining lymph nodes, and sorted on FACSAria by the expression of CXCR5, PD-1 and BCL6-YFP. Naive CD4+ T cells were CD4+ CD44lo CD62Lhi cells from unimmunized mice.

Glycopeptide glycoforms analysis of mouse transferrin

Project description:Collagen type II-induced arthritis (CIA) is a disease, which includes inflammatory and autoimmune reactions. Spleen protein extracts were subjected to 2D-DiGE and MS-MALDI-TOF/TOF analysis to identify proteins that were different in concentration ally expressed in CD38-KO and B6 WT mice with arthritis or with inflammation. Using multivariate analyses, in Col-II-immunized mice 20 differentially expressed spleen protein species were able to discriminate between WT and CD38-KO mice. Among them, multiple serotransferrin (Tf) species were identified. In contrast, in CFA/IFA-treated mice, the distinct protein profile, which discriminates between CD38-KO and WT mice, was unrelated with Tf. Unexpectedly, non-immunized CD38-KO mice showed a distinct proteome profile as compared with that in non-immunized WT mice, and again multiple protein species were identified as Tf. By using a LC-TOF-MS method to separate and detect Tf glycopeptide glycoforms in serum samples, differences in the Tf glycosylation pattern between CD38-KO and WT mice sera were found in both non-immunized and CIA+ mice. Data on Tf spots separated by 2-DE indicate differences in glycosylation related with NeuGc content. In summary, Tf changed significantly in its glycosylation pattern in mice with arthritis

Project description:Collagen type II-induced arthritis (CIA) is an autoimmune disease that is accompanied by a complex host systemic response, which includes inflammatory and autoimmune reactions. Since to develop CIA is required the injection of antigen in complete Freund’s adjuvant (CFA), which is a water-in-mineral-oil emulsion containing killed Mycobacteria, systemic response proteins related with inflammation can confound the detection or diagnosis of arthritis. CIA in CD38 deficient mice (CD38 KO) is milder than that in C57BL/6 (B6 WT) mice. Protein extracts from spleen were subjected to 2D-DiGE and MS-MALDI-TOF/TOF analysis to identify proteins that were differentially expressed in CD38 KO and B6 WT mice with arthritis, with inflammation, or non-immunized. Differential protein expression was validated by ELISA, or Western-blotting.
