Project description:IL-17 Inhibition in Spondyloarthritis Associates with Subclinical Gut Microbiome Perturbations and a Distinctive IL-25-Driven Intestinal Inflammation Volatile short-chain fatty acids analysis. Approximately 50 mg of sample were placed in 2 ml borosilicate vials capped with a screw cap with silicone septum. The capped vials were stored at -20 C and thawed immediately prior to analysis. The GC-MS analysis was carried out using the Agilent 7200 GC/QTOF equipped with robotic sampler system. The volatiles from the sample were extracted from headspace using Polydimethylsiloxane/Divinylbenzene (PDMS/DVB) df 65 um Solid Phase Microextraction Fiber (SPME) for 30 minutes at 50 C. The fiber was then inserted into the injector equipped with Merlin septum heated to 250 C and the adsorbed compounds were desorbed for 1 minute.

Matrix Elasticity Does Not Affect Replicative Senescence or DNA Methylation Patterns of Mesenchymal Stem Cells [gene expression profiling]

Project description:Matrix elasticity influences differentiation of mesenchymal stem cells (MSCs) but it is unclear if these effects are only transient - while the cells reside on the substrate - or if they reflect persistent lineage commitment. In this study, MSCs were continuously culture-expanded in parallel either on polydimethylsiloxane (PDMS) gels of different elasticity or on tissue culture plastic (TCP) to compare impact on replicative senescence, in vitro differentiation, gene expression, and DNA methylation (DNAm) profiles. The maximal number of cumulative population doublings was not affected by matrix elasticity. Differentiation towards adipogenic and osteogenic lineage was increased on soft and rigid biomaterials, respectively - but this propensity was no more evident if cells were transferred to TCP. Global gene expression profiles and DNAm profiles revealed relatively few differences in MSCs cultured on soft or rigid matrices. Furthermore, only moderate DNAm changes were observed upon culture on very soft hydrogels of human platelet lysate. Our results support the notion that matrix elasticity influences cellular differentiation while the cells are organized on the substrate, but it does not have major impact on cell-intrinsic lineage determination, replicative senescence or DNAm patterns. MSCs cultivated either on polydimethylsiloxane (PDMS) gels of different elasticity or on tissue culture plastic (TCP) to compare impact on gene expression profiles.

Project description:Samples from extraction of volatile organic compounds from frogs using two sampling methods: in vivo and sampling from the skin and a DVB/CAR/PDMS-SPME fiber. Splitless injection mode using a desorption temperature of 250C. Separation was performed initiating to 40C for 3 min, and then increasing the temperature to 100C using a rate of 6C/min, afterwards incresing to 200C changing the rate to 4C/min, and finally increasing to 300C, using a rate of 20C/min, and maintaing to this temperature for 3 min. Analysis were performed in a GC HP 6890 Series equipped with an Agilent Mass Selective Detector 5973 (Agilent technologies, Palo Alto, CA, USA). Separation was performed on a BP-5 capillary GC column (30 m 0.25 mm 0.25 um, SGE, Austin, TX, USA) using helium as a carrier gas at a flow rate of 1.0 mL/min.

Substrate elasticity does not impact on DNA methylation changes during differentiation of pluripotent stem cells.

Project description:Substrate elasticity may direct cell-fate decisions of stem cells. However, it is largely unclear how matrix stiffness impacts on differentiation of induced pluripotent stem cells (iPSCs) and if this is also reflected by epigenetic modifications. We have therefore cultured iPSCs on tissue culture plastic (TCP) and polydimethylsiloxane (PDMS) with different Young´s modulus (0.2 kPa, 16 kPa, or 64 kPa) to investigate the sequel on growth and differentiation towards endoderm, mesoderm, and ectoderm. Immunofluorescence and gene expression of canonical differentiation markers was hardly affected by the substrates. Notably, when we analyzed DNA methylation profiles of iPSCs or after three-lineage differentiation, we did not see any significant differences on the three different PDMS elasticities. Only when we compared DNA methylation profiles on PDMS-substrates versus TCP, we observed clear epigenetic differences, particularly upon mesodermal differentiation. Taken together, stiffness of PDMS-substrates did not impact on directed differentiation of iPSCs, whereas the moderate epigenetic differences on TCP might also be attributed to other chemical parameters.

A global proteomic approach was used in order to identify the most abundant proteins for the comparison between E. coli adhesion in PDMS 9 kPa and 574kPa, (polydimethylsiloxane,), and between HA 44Pa and 2kPa (hyaluronic acid gels).

Modular automated microfluidic cell culture platform reduces glycolytic stress in cerebral cortex organoids

Project description:Organ-on-a-chip systems combine microfluidics, cell biology, and tissue engineering to culture 3D organ-specific in vitro models that recapitulate the biology and physiology of their in vivo counterparts. Here, we have developed a multiplex platform that automates the culture of individual organoids in isolated microenvironments at user-defined media flow rates. Programmable workflows allow the use of multiple reagent reservoirs that may be applied to direct differentiation, study temporal variables, and grow cultures long term. Novel techniques in polydimethylsiloxane (PDMS) chip fabrication are described here that enable features on the upper and lower planes of a single PDMS substrate. RNA sequencing (RNA-seq) analysis of automated cerebral cortex organoid cultures shows benefits in reducing glycolytic and endoplasmic reticulum stress compared to conventional in vitro cell cultures.
