Dietary lipids accumulate in macrophages and stromal cells and change the microarchitecture of the mesenteric lymph node II

Project description:Obesity has emerged as a worldwide problem in human health. Dietary lipids are taken up and transported via lymphatics into the circulatory system. During this process, lipids pass the mesenteric lymph node (mLN). These organs filter the lymph fluid for foreign antigens to induce and control immune responses. Alteration of that function during obesity has only slightly been studied. Here, we characterize changes within the microarchitecture of the mLN during high levels of lipid transport and highlight the role of stromal cells. Microarray experiments detected gene probes expressed by mLN stromal cells. Transmission electron microscopy enabled us to identify lipid droplets in different stromal cells, and in macrophages. Sizes, numbers and intercellular distances increased after 10 weeks of a high-fat diet. Thus, we propose that changes in the microarchitecture and increased accumulation of lipid droplets in stromal cells and macrophages have an influence on the immunological function of the mLN.

Project description:All free-living microorganisms homeostatically maintain the fluidity of their membranes by adapting lipid composition to environmental temperatures. A quantitative description of how organisms maintain constant fluidity at all growth temperatures has not been achieved. By quantifying both enzymes and metabolic intermediates of the Escherichia coli fatty acid and phospholipid synthesis pathways, we discover how E. coli measures steady-state temperature and restores optimal membrane fluidity within a single generation after temperature shocks.

Hypometabolism to survive the long polar night in the diatom Fragilariopsis cylindrus

Project description:Diatoms, the major eukaryotic phytoplankton in polar regions, are essential to sustain food webs. As such, it is fundamental to understand the physiological mechanisms and associated molecular basis of resilience to polar night of diatoms. Here, we report an integrative approach that reveals that in prolonged darkness, the cell enters a state of hypometabolism associated with reduced transcriptional activity during which no cell division occurs. Minimal energy is provided by respiration via alternative oxidase and progressive degradation of protein, carbohydrate and lipid stores. We also report internal structural changes that manifest the morphological acclimation of cells to darkness. Our results further indicate that immediately after returning to light, the majority of cells were able to use photoprotective mechanisms and resume photosynthesis. Divisions of surviving cells resumed at rates similar to those before darkness. Our study demonstrates the robustness of this species to prolonged darkness at low temperatures.

Transcriptional impacts of lipids on hESC

Project description:Pluripotent stem cells can be maintained in a continuum of cellular states with distinct features. Exogenous lipid supplements are commonly utilized to shift the balance of global metabolism relieving the dependence on de novo lipogenesis. However, it is largely unexplored how specific lipid components regulate metabolism and pluripotency state. In this study, we investigate the impact of lipid supplements on human embryonic stem cells (hESCs), and report that signaling lipid lysophosphatidic acid (LPA) is the key component to shift the metabolic landscape in lipid supplement AlbuMAX. Although the maintenance of ERK phosphorylation and primed pluripotency is independent of exogenous lipids, LPA increases ERK phosphorylation, especially upon niche disintegration. We further demonstrate that LPA leads to distinctive transcriptome profile that is not associated with de novo lipogenesis. We also show that LPA causes unique and reversible phenotypes in cell cycle, morphology and mitochondria. This study reveals an LPA -induced primed state that allow people to drastically alter cell physiology for basic research and stem cell applications with hESCs.
