Mytilus galloprovincialis, sperm, LC-MSMS

ABSTRACT: Sperm contains essential proteins for interaction with eggs, however, there are only several sperm proteins reported with important role in fertilization, and gamete proteomics are limited in marine invertebrate species. We present here a sperm proteomic profile of marine mussel Mytilus galloprovincialis. There are 816 proteins were successfully identified by LC-MS/MS based on 1-DE SDS-PAGE. Many of the identifications are relevant to sperm cell physiology and mtDNA functioning. The results will contribute to better understand the proteins involved in fertilization in M. galloprovincialis, as well as the other marine invertebrate species.

Proteomic analysis of Dpy19l2-deficient human globozoospermia reveals multiple molecular defects

Project description:Globozoospermia is a severe teratozoospermia characterized by round-headed spermatozoa without acrosomes, resulting in male infertility. The clinical treatment of globozoospermia involves primarily intracytoplasmic sperm injection because of the low fertilization rate. The deletion of Dpy19l2 is known to be the major cause of globozoospermia. The defects in Dpy19l2-deficient human sperm at the protein level were characterized by performing a isotope-based quantitative proteomic analysis between Dpy19l2-deficient globozoospermic spermatozoa and normal controls. A total of 2,549 proteins were identified, including 495 differentially expressed proteins (fold-change >2), of which 322 were upregulated and 123 were downregulated. The levels of several important proteins, including SPACA 1, IZUMO1, ZPBP1, and PLCZ1, were decreased in globozoospermic sperm. Bioinformatics analysis indicated the Dpy19l2-deficient sperm presented molecular defects in acrosome, chromatin, sperm-egg interaction, and fertilization. This study may provide insights into the mechanism of globozoospermia and help develop better treatments to increase the success rate of fertilization.

Combined sperm proteome of the monarch butterfly Danaus plexippus

Project description:We characterized sperm from the seminal vesicles of male monarch butterflies (Danaus plexippus), in triplicate, identifying 548 high confidence proteins. As with all but the most basal lepidopteran species male monarch butterflies are sperm heteromorphic, producing fertilization competent and anucleate fertilization incompetent sperm morphs. Comparing this data to the sperm proteomes of the Carolina sphinx moth (Manduca sexta) and the fruit fly (Drosophila melanogaster) demonstrated high levels of functional coherence across proteomes, and conservation at the level of protein abundance and post-translational modification within Lepidoptera. Comparative genomic analyses revealed a significant reduction in orthology among Monarch sperm genes relative to the remainder of the genome in non-Lepidopteran insects. A substantial number of sperm proteins were found to be specific to Lepidoptera, lacking detectable homology outside this taxa. These findings are consistent with a burst of genetic novelty in the sperm proteome concurrent with the origin of heteromorphic spermatogenesis early in Lepidoptera evolution.

Subcellular fractionation of boar spermatozoa provides a novel view of the perinuclear theca proteome

Project description:The perinuclear theca (PT) is a highly condensed, largely insoluble protein structure that surrounds the nucleus of eutherian spermatozoa. While this structure is known to be important for fertilization, little is known regarding its proteomic composition. Recent reports have indicated that the PT unexpectedly houses several somatic proteins, such as core histones, which may be important post-fertilization during re-modelling of the male pronucleus. To explore the proteomic composition of the PT we performed the first in depth, label-free proteomic characterization of the PT of boar spermatozoa. To do this, a unique subcellular fractionation protocol was first performed to isolate the PT and increase our ability to detect lowly abundant sperm proteins. Through the use of this subcellular fractionation technique we were able to quantify 1802 proteins, a result that represents unparalleled depth of coverage for the boar sperm proteome and exceeds the entire known proteome of the Sus scrofa species so far. In the PT structure itself we identified 813 proteins and confirmed the presence of previously characterized PT proteins including the core histones H2A, H2B, H3 and H4, as well as Ras-related protein Rab-2A (RAB2A) and Rab-2B (RAB2B) amongst other RAB proteins. In addition to these previously characterized PT proteins, our data also revealed that the PT is replete in proteins critical for sperm-egg fusion including: Izumo sperm-egg fusion proteins 1-4 (IZUMO1-4) and phosphoinositide phospholipase C (PLCZ1).

A Restricted Role for Sperm-borne MicroRNAs in Mammalian Fertilization

Project description:Prototypical micro RNAs (miRNAs) are 21~25-base-pair RNAs that regulate differentiation, carcinogenesis and pluripotency by eliminating mRNAs or blocking their translation, processes collectively termed RNA interference (RNAi). RNAi mediated by miRNAs regulates early development in zebrafish, and mouse embryos lacking the miRNA precursor processor, Dicer, are inviable. However, the role of miRNAs during mammalian fertilization is unknown. We here show using microarrays that miRNAs are present in mouse sperm structures that enter the oocyte at fertilization. Sperm contained a broad profile of miRNAs and a subset of potential mRNA targets were expressed in fertilizable, metaphase II (mII) oocytes. Oocytes contained transcripts for the RNAinduced silencing complex (RISC) catalytic subunit, EIF2C3 (formerly AGO3). However, levels of sperm-borne miRNA (measured by quantitative PCR) were apparently low relative to those of unfertilized, mII oocytes, and fertilization did not alter the part of the mII oocyte miRNA landscape that included the most abundant sperm-borne miRNAs. Coinjection of mII oocytes with sperm heads plus anti-miRNAs - to suppress miRNA function - did not perturb pronuclear activation or preimplantation development. Contrastingly, we provide evidence that nuclear transfer by microinjection alters the miRNA profile of enucleated oocytes. These data argue that sperm-borne prototypical miRNAs play a limited role, if any, in mammalian fertilization or early preimplantation development. Proteomic changes in human sperm during sequential in vitro capacitation and acrosome reaction.

Project description:The male gamete is not completely mature after ejaculation and requires further events in the female genital tract to acquire fertilizing ability, including the processes of capacitation and acrosome reaction. In order to shed light on dynamic protein changes experienced by the sperm cell in preparation for fertilization, a comprehensive quantitative proteomic profiling based on isotopic peptide labeling and liquid chromatography followed by tandem mass spectrometry was performed on spermatozoa from three donors of proven fertility under three sequential conditions: purification with density gradient centrifugation, incubation with capacitation medium, and induction of acrosome reaction by the exposure to the calcium ionophore A23187. After applying strict selection criteria for peptide quantification and for statistical analyses, 36 proteins with significant changes in their relative abundance within sperm protein extracts were detected. Moreover, the presence of peptide residues potentially harboring sites for post-translational modification was revealed, suggesting that protein modification may be an important mechanism in sperm maturation. In this regard, increased levels of proteins mainly involved in motility and signaling, both regulated by protein modifiers, were detected in sperm lysates following incubation with capacitation medium. In contrast, less abundant proteins in acrosome-reacted cell lysates did not contain potentially modifiable residues, suggesting the possibility that all those proteins might be relocated or released during the process. Protein-protein interaction analysis revealed a subset of proteins potentially involved in sperm maturation, including ERLIN2, GGH, TMED 10, and ATP synthases. These results contribute to the current knowledge of the molecular basis of human fertilization and propose new candidates to be validated as modulators of male fertility.

Molecular evolution of sperm competition in mice

Project description:Sperm are amongst the most rapidly evolving cell types due, in part, to their central role in post-mating competitive reproductive success (sperm competition) and participation in coevolving male-female interactions. To establish the molecular basis of sperm divergence we have conducted a whole-cell, semi-quantitative proteomic approach to characterize sperm divergence in three closely related Mus species (musculus, spretus and spicilegus), which experience different regimes of sperm competition and exhibit remarkable variation in sperm production, energetics, motility and fertilization capacity.

Differential proteomics of human seminal fluid Normo Vs Astehnozoospermia

Project description:Scarcely understood defects lead to asthenozoospermia which results in poor fertility outcomes. Incomplete knowledge of these defects hinders the development of new therapies and reliance on interventional therapies, such as in vitro fertilization, increases. Sperm cells, being transcriptionally and translationally silent, necessitate the proteomic approach to study the sperm funnction. We have performed a differential proteomics analysis of human sperm and seminal fluid and identified over 1,700 proteins. We have included 667 proteins in sperm and 430 proteins in seminal plasma dataset for further analysis. Statistical and mathematical analysis combined with pathway analysis and self organizing maps clustering and correlation was performed on the dataset. It was found that sperm proteomic signature combined with statistical analysis as opposed to the seminal plasma proteomic signature can differentiate the normozoospermic versus the asthenozoospermic sperm samples.
