Characterization of the phospho-signalling mediated by the sodium-potassium ATPase in response to ouabain

ABSTRACT: In this project we applied a workflow for phosphoproteomics analysis to characterize the phosphorylation signalling that is triggered by the cardiotonic steroid ouabain. Ouabain is a ligand for the sodium-potassium ATPase (NKA). Low concentration of ouabain trigger oscillation of intracellular calcium concentration and modulate the phosphorylation of several signaling molecules, impacting cellular proliferation, apoptosis and cell-to-cell contacts. Monkey kidney cells (COS-7) were either treated for 10 or 20 minutes with 100 nM ouabain, or left untreated. We identify 15,348 phospho-sites, of which 1,941 are significantly regulated at either one of the time points employed. Analysis of the significantly regulated phospho-sites shed light on the molecular mechanisms by which ouabain affects cell junctions, proliferation and apoptosis.

Project description:Tartrate-resistant acid phosphatase (TRAP/ACP5) is a metalloenzyme of the group of acid phosphatases. High levels of TRAP protein have been associated with the development of cancer metastasis and with clinically relevant parameters of cancer progression and cancer aggressiveness. In order to investigate the molecular pathways responsible for the role of TRAP in such cancer related processes, we examined the changes in the proteome and phosphoproteome of MDA-MB-231 breast cancer cells determined by TRAP protein overexpression or knock-down. The dataset contains two different experimental setups. The first setup consists of the comparison between MDA-MB-231 breast cancer cells stably transfected with a full-length rat TRAP (TRAP3high) and MDA-MB-231 cells transfected with a mock insert (ctrl). The second setup consists of the comparison between TRAP3high cells transfected with a scrambled shRNA (scr) and TRAP3high cells transfected with either of two shRNAs targeting TRAP (sh2 and sh3+4). In the first experimental setup, we performed quantitative phosphoproteomic and proteomic analyses using SILAC. The two analyses identified respectively 3,290 unique phosphorylation sites corresponding to 1,059 genes, and 7,957 proteins corresponding to 7,846 genes. In the second experimental setup, we performed quantitative proteomics analysis using TMT, identifying 9,848 proteins corresponding to 9,189 genes. Integrated analysis of the generated data revealed a regulation of proteins involved in the cell adhesion process and extracellular matrix organization network. In particular, TGFβ isoform 2 (TGFβ2), TGFβ receptor type 1 (TβR1) and SMAD2 protein levels were increased upon TRAP upregulation, as well as phosphorylation of CD44 on residues localized in the intracellular portion of the protein. Finally, these results were validated by functional blocking and chemical inhibition of TGFβ2/TβR, which decreased TRAP-dependent cell migration and proliferation, and by blocking of CD44, which reduced TRAP3high cell proliferation. Altogether, we show that TRAP promotes metastasis-related cell properties in breast cancer cells via TGFβ2/TβR and CD44, thereby identifying a potential signaling mechanism associated to TRAP action in breast cancer cells.

Project description:Part I - SI-NET tumors High Resolution Isoelectric Focusing (HiRIEF) LC-MS and relative quantification by iTRAQ 8-plex was used to analyze 14 small intestinal neuroendocrine tumors (SI-NETs). The data was obtained from two separate TMT10plex sets and linked by a single internal pooled standard. The data was obtained from two separate TMT 10-plex experiments. TMT set1 includes a time course experiment upon pevonedistat treatment of CNDT2 cells with harvests at 2h, 6h, 12h and 24h after treatment as well as of untreated control cells. Isobaric tag labelling of peptide samples with TMT10plex was used as follows. Biological duplicate controls (TMT channels 126, 127N), duplicate 2h (127C, 128N), duplicate 6h (128C, 129N), duplicate 12h (129C, 130N) and duplicate 24h (130C, 131) samples were employed. TMT set2 includes a time course experiment upon pevonedistat treatment of HC45 cells with harvests at 2h, 6h, 12h and 24h after treatment as well as of untreated control cells. Isobaric tag labelling of peptide samples with TMT10plex was used as follows. Project description:Spermatophyte pollen tubes and root hairs have been used as single-cell-type model systems toward understanding the molecular processes underlying polar growth of plant cells. Horsetail (Equisetum arvense L.) is a perennial herb species in Equisetopsida, which creates separately growing spring and summer stems in its life cycle. The mature chlorophyllous spores produced from spring stems can germinate without dormancy. Here we report the cellular features and protein expression patterns in five stages of horsetail spore germination (mature spores, rehydrated spores, double-celled spores, germinated spores, and spores with protonemal cells). Using 2-DE combined with mass spectrometry, 80 proteins were found to be differentially expressed upon spore germination. Among them, proteins involved in photosynthesis, protein turnover, and energy supply were over-represented. Thirteen proteins appeared as isoforms on the gels, indicating the potential importance of post-translational modification. In addition, the dynamic changes of ascorbate peroxidase, peroxiredoxin, and dehydroascorbate reductase implied that reactive oxygen species homeostasis is critical in regulating cell division and tip-growth. The time course of germination and diverse expression patterns of proteins in photosynthesis, energy supply, lipid and amino acid metabolism indicated that heterotrophic and autotrophic metabolism were necessary in light-dependent germination of the spores. Twenty-six proteins were involved in protein synthesis, folding, and degradation, indicating that protein turnover is vital to spore germination and rhizoid tip-growth. Furthermore, the altered abundance of small G protein Ran1, 14-3-3 protein, actin, and Caffeoyl-CoA O-methyltransferase revealed that signaling transduction, vesicle trafficking, cytoskeleton dynamics, and cell wall modulation were critical to cell division and polar growth. These findings provide up-to-date evidences for understanding fern spore asymmetric division and rhizoid polar growth.
