LC-MS/MS after Immunoprecipitation with Mbd5 in zebrafish at 4dpf

ABSTRACT: To address how Mbd5 binding to m5C RNAs mediates transcriptional activation of its target genes. Using the Tg[zhsp70l:FLAGmbd5iso2-E2AGFP] zebrafish, we performed heat shock at multiple developmental stages and carried out co-immunoprecipitation (co-IP) at 4 dpf using the FLAG antibody, followed by mass spectrometry

Complex biological processes are regulated by both genetic and epigenetic programs. One class of epigenetic modifications is methylation. Evolutionarily conserved methyl-CpG-binding domain (MBD)-containing proteins are known as readers of DNA methylation. MBD5 is linked to multiple human diseases but its mechanism of action remains unclear. Here we report that the zebrafish Mbd5 does not bind to methylated DNA; but rather, it directly binds to 5-methylcytosine (m5C)-modified mRNAs and regulates  ...[more]
