Project description:Spontaneously hypertensive rat (SHR; Rat Genome Database (RGD) ID: 631848) is probably the most commonly used rodent model for essential hypertension. Its minimal congenic subline PD5 ( SHR.PD-(D8Rat42-D8Arb23)/Cub; (RGD ID: 1641851)) which differs by a short segment of chromosome 8 (encompassing only 7 genes) displays lower levels of blood pressure and amelioration of cardiac fibrosis compared to SHR. We used microarray aproach to unravel gene expression differences between SHR and PD5 responsible for the different phenotypes in our rat strains.

Project description:The dissection of genetic components of metabolic syndrome is currently one of the most important research topics. Plzf gene is a promising candidate gene. Previously, we described minimal congenic strain PD5 derived from spontaneously hypertensive rat (SHR), containing congenic segment of polydactylous (PD/Cub) origin with a 2964bp intronic deletion within Plzf gene and displaying attenuated features of metabolic syndrome. To unravel the genetic background responsible for the observed of this phenotype in PD5 we compared liver transcriptomes and physiologic parameters between SHR and PD5 strains without intervention and after dexamethasone administration.

Expression data was determined for 2 ages: 6 week and 24 week with whole kidney RNA.

Project description:This study examined the renal gene expression profiles between spontaneously-hypertensive rats (SHR) and Wistar-Kyoto (WKY) rats at a pre-hypertensive stage (3 weeks of age) and hypertensive stage (9 weeks of age). In addition, age-related changes in gene expression patterns were examined from 3 to 9 weeks in both WKY and SHR.
