Project description:In order to screen and identify biomineralization gene, microarray technique was used to reveal tissues specific expression genes in the brunet mantle edge (ME), mantle centre (MC), and both ME and MC (ME-MC) from assembled transcriptome contigs of Pinctada fucata martensii, ideal pearl oyster for the study of biomineralization. Tissues of ME, MC, hepatopancreas, foot, gill, adductor muscle, heart and intestine were sampled from two females and one male pearl oyster. Gonad was sampled from above three individuals and another two male and one female. Equal amount RNA of each individual hepatopancreas, foot, gill, adductor muscle, heart and intestine were mixed as a composite viscera sample (CV), and equal amount gonad RNA from one male and one female were combined together as a gonad sample (GS). Hybridizations were performed with twelve samples of ME, MC, CV and GS. Gene expression in ME, MC, gonad and other tissues were measured. Gonads were sampled from 6 individuals, and other tissues were sampled from above three individuals.

Project description:The appearance of hard mineralized exoskeletons is a critical leap for animal evolution and partially lead to the explosion of diverse animals during the Cambrian, for example, molluscs. A majority of molluscs have mineralized shells to protect themselves. Despite numerous studies that have studied the remarkable mechanical properties of shells, the origin of shell formation is still elusive. Hence, this study investigated the overlooked shell proteome of chitons, which belong to polyplacophoran, Aculifera of Mollusca. By comparing the shell proteome to well-studied Conchifera groups, we inferred possible ancestral biomineralization toolkits of stem-group Mollusca. Taking advantage of the recently sequenced chiton mantle transcriptome and genome, eight core biomineralization proteins were identified by proteomics. Surprisingly, in contrast to previous thought that shell formation is convergently evolved, two important shell matrix proteins, Nacrein-like and Pif-like proteins were found to be conserved among Aculifera and Conchifera groups. Our findings identify a missed link of mineralized shell evolution in Mollusca and pose a hypothesis that stem-group molluscs have already evolved core biomineralization toolkits, which likely facilitate the formation of mineralized shells for protection that partially leads to their explosion.

Project description:Mussels synthesize an interesting class of biological materials with unique properties to adhere onto virtually any solid surface. Inter- and cross-species analyses have revealed that mussel byssus fabrication is influenced by environmental factors. In this study, proteins expressed specifically byssus and in the byssal-producing organ (foot) were examined for two mussel species from the Mytilidae family using liquid chromatography coupled with high-resolution tandem mass spectrometry (LC-MS/MS). The main goal was to describe which proteins are exclusively expressed in the mussel's foot gland, which is responsible for byssus secretion. Proteins uniquely found in the foot samples have been found by comparing to a control tissue (mantle). The overlap with proteins found in byssus samples were also investigated. A restricted list of these proteins have been highlighted as byssus-related, believed to contribute to the unique properties of byssus, such as securing adhesion and toughness, and plasticity based on environmental factors (including promoter regulation). This work may provide further directions on thread biofabrication and additional information on its regulation, as well as contribute to a more complete description of the mussel proteome.

Project description:Sepioloidea lineolata, the striped pyjama squid (family Sepidariidae), is a small species of benthic squid distributed along the Southern Indo-Pacfic coast of Australia. All Sepiadariid squids are known to secrete large volumes of viscous slime when stressed. The proteome of the slime, dorsal and ventral mantle muscle, the dorsal and ventral mantle epithelium and ventral mantle glands was analysed by combining label-free quantitative analysis using high resolution mass spectrometry data with an S. lineolata transcriptome assembled from give tissues including slime. A total of 28 highly positively differentially expressed proteins were identified within the slime and were predominately comprised of a host of enzymes including peptidases and protease inhibitors. Seven of these proteins contained predicted signal peptides, indicating classical secretion, with four proteins having no identifiable domains or similarity to any known proteins.
