Project description:We conducted an RNA-sequencing on the placentomes that were collected from 4 and 3 cows with and without retained placenta, respectively. Placentomes were collected from the cows within 30 minutes after parturition and samples were taken from the feto-maternal junction sites of the placentomes. These samples were used for RNA-sequencing and the results showed 240 genes that were differentially expressed between the two groups. These genes could be candidate genes that are involved in the development of placental retention in cattle.

Biomarkers for diagnosing the retained placenta in cows-does the analysis of serum proteome keep the secret?

Project description:Retained placenta (RP), a quite common disorder in dairy cows, shows a high negative impact on their health status and milk production. Therefore, the biomarkers indicating the risk for RP could yield benefits in dairy management. The study aimed to investigate the serum proteome as the source of the biomarkers for diagnosing the RP. Using the high-resolution liquid chromatography-tandem mass spectrometry, we compared the composition of serum proteome between 9 cows with the RP and 6 with the physiologic puerperium (PP). Among 651 identified proteins, seven had higher, and three showed lower abundance in RP than in the PP group. The differently abundant proteins participated in 15 pathways: six related to hemostasis, three involved in the lipoprotein metabolism, and the remaining ones associated with redox homeostasis, post-translational modification, scavenging, etc. Validation of the proteomic results, using ELISA and spectrophotometric assays, showed that the two identified proteins, haptoglobin, and lipopolysaccharide-binding protein, could represent reliable RP biomarkers. Our results confirmed that the analysis of serum proteome shed "the light" on the counterparty piece(s) of the RP pathogenesis "puzzle" and could be an efficient "tool" in mining for RP biomarkers.

Effect of senktide administration on the embryo collected from superovulated cows

Project description:To clarify the effect of intravenous senktide administration on the quality and developmental stage of collected embryo from superovulated cows, mRNA expression profiles of the embryo were investigated. Hierarchical cluster analysis with the expression levels of all genes was divided these cows into three clusters. First cluster was composed of control cows, second cluster contained senktide (30 nmol/min, 2h) treated cows and third cluster consisted of senktide (300 nmol/min, 2h) treated cows.

Transcriptome analysis of the endometrium, placenta, and liver in lactating and non-lactating dairy cows

Project description:Infertility in lactating dairy cows is explained partially by the metabolic state associated with high milk production. The hypothesis was that lactating and non-lactating cows would differ in endometrial and placental transcriptomes during early pregnancy (day 28 to 42) and this difference would explain the predisposition for lactating cows to have embryonic loss at that time. Cows were either milked or not milked after calving. Reproductive [endometrium (caruncular and intercarunclar) and placenta] and liver tissues were collected on day 28, 35, and 42 of pregnancy. The primary hypothesis was rejected because no effect of lactation on mRNA abundance within reproductive tissues was found. Large differences within liver demonstrated the utility of the model to test an effect of lactation on tissue gene expression. Major changes in gene expression in reproductive tissues across time were found. Greater activation of the transcriptome for the recruitment and activation of macrophages was found in the endometrium and placenta. Changes in glucose metabolism between day 28 and 42 included greater mRNA abundance of rate-limiting genes for gluconeogenesis in intercaruncular endometrium and evidence for the establishment of aerobic glycolysis (Warburg effect) in the placenta. Temporal changes were predicted to be controlled by CSF1, PDGFB, and JUN. Production of nitric oxide and reactive oxygen species by macrophages was a mechanism to promote angiogenesis in the endometrium. Reported differences in pregnancy development for lactating versus non-lactating cows could be explained by systemic glucose availability to the conceptus and appear to be independent of the endometrial and placental transcriptomes.

Plasma Metabolomics Reveals Pathogenesis of Retained Placenta in Dairy Cows.

Project description:The complex etiology and pathogenesis of retained placenta (RP) bring huge challenges for researchers and clinical veterinarians in investigating the pathogenesis and treatment schedule. This study aims to investigate the pathogenesis of RP in dairy cows by plasma metabolomics. As subjects, 10 dairy cows with RP and 10 healthy dairy cows were enrolled according to strict enrollment criteria. Imbalanced antioxidant capacity, reduced Th1/Th2 cytokine ratio, and deregulation of total bilirubin (T-bil), alkaline phosphatase (ALP), and reproductive hormones were shown in dairy cows with RP by detecting biochemical indicators, oxidation and antioxidant markers, and cytokines in serum. Plasma metabolites were detected and analyzed by a liquid chromatography-mass spectrometry (LC-MS) system coupled with multivariate statistical analysis software. A total of 23 potential biomarkers were uncovered in the plasma of dairy cows with RP. The metabolic pathways involved in these potential biomarkers are interconnected, and the conversion, utilization, and excretion of nitrogen were disturbed in dairy cows with RP. Moreover, these potential biomarkers are involved in the regulation of antioxidant capacity, inflammation, and autocrine or paracrine hormone. All of these findings suggest that an imbalance of these potential biomarkers might be responsible for the imbalanced antioxidant capacity, reduced Th1/Th2 cytokine ratio, and deregulation of reproductive hormones in dairy cows with RP. The regulation of metabolic pathways involved in these potential biomarkers represents a promising therapeutic strategy for RP.
