The normal duct-lobular system of the breast is lined by two epithelial cell types, inner luminal secretory cells and outer contractile myoepithelial cells. We have generated comprehensive expression profiles of the two normal cell types, using immunomagnetic cell separation and gene expression microarray analysis. Project description:Three apoptosis inducing proteins, ACO1, STK3 and XBP1 were discovered in a previous reverse transfection screen. HEK293T cells were harvested and RNA extracted from the cells after over-expression of these 3 proteins at 12, 24 and 48 hours. A positive apoptosis inducer staurosporine and a negative control transfection reagent only were also included at each time point. Each sample had an experimental replicate.

Project description:Comparing Vg9Vd2 T cells with CD8 ab T cells To compare the responses of resting and activated human Vg9Vd2 T cells (mainly CD4-CD8-) to those of conventional CD8+ ab T cells, we used Hver2.1.1 microarrays containing ~15,000 cDNAs. CD8 T cells were chosen as the conventional counterpart of human Vg9Vd2+ cells because the biology and clinical application of Vg9Vd2 T cells is most often rationalized in the context of the cells proven cytolytic potential. This does not withstand the fact that both conventional CD8 T cells and Vg9Vd2+ cells share with T-helper cells and other cells the capacity to express various cytokines and chemokines.

Transcription profiling of Arabidopsis cells to iron stress

Project description:After 7 days under normal culture conditions, an iron stress was imposed by addition of iron citrate (500 uM final concentration). This time constituted the time zero (T0) of the time-course experiment. Cells were collected from separate Erlenmeyer flasks at times T0, T+5 min, T+15 min, T+30 min, T+60 min, T+6 h, and T+24h. After filtration, cells were washed with water twice, frozen in liquid nitrogen and stored at 80C before processing for RNA extraction.
