Methylation profiling of human placenta villi obtained at 7-8 and 13-14 weeks gestational age

ABSTRACT: The human intraplacental environment undergoes major changes at the end of the first trimester associated with onset of the maternal blood flow. We collected placental villi samples before and after maternal blood flow to profile methylation changes at this crucial time window.

Reducing cell intrinsic immune responses to mRNA vaccine constructs improves adaptive immune response in mice

Project description:The response to mRNA vaccines needs to be sufficient for immune cell activation and recruitment but moderate enough to ensure efficacious antigen expression. The choice of the cap structure and use of N1-methylpseudouridine (m1Ψ) instead of uridine, which have been shown to reduce RNA sensing by the cellular innate immune system, has led to improved efficacy of mRNA vaccine platforms. Understanding how RNA modifications influence the cell intrinsic immune response may help in the development of more effective mRNA vaccines. In the current study, we compared mRNA vaccines in mice against influenza virus using three different mRNA formats: uridine-containing mRNA (D1-uRNA), m1Ψ- modified mRNA (D1-modRNA), and m1Ψ-modified mRNA with a cap1 structure (cC1-modRNA). D1-uRNA vaccine induced a significantly different gene expression profile to the modified mRNA vaccines, with an upregulation of Stat1 and RnaseL, and increased systemic inflammation. This correlated with a significantly reduced antigen specific antibody responses and reduced protection against influenza virus infection compared to D1-modRNA and cC1-modRNA. Incorporation of m1Ψ alone without cap1 improved antibodies, but both modifications were required for the optimum response. Therefore, the incorporation of m1Ψ and cap1 alters protective immunity from mRNA vaccines by altering the innate immune response to the vaccine material

Illumina methylation EPIC arrays of human blood at baseline and after 18 months of dietary interventions

Project description:To highlight molecular mechanisms underlying the beneficial metabolic effects of a Mediterranean (MED) diet, a MED diet enriched in polyphenols and reduced in red/processed meat (greenMED), and a diet with healthy dietary guidelines (HDG), we analyzed their effects on the methylome in the 18-month DIRECT PLUS randomized controlled trial. The methylation arrays contains individuals from the three intervention arms (MED, greenMED, HDG) for baseline (T0). We will gladly provide further metadata upon request.

Transcription profiling of mouse blood from native and Plasmodium berghei induced experimental cerebral malaria untreated or treated with activated charcoal.

Project description:We found that oral activated charcoal (oAC) provided significant protection against P. berghei ANKA-induced ECM, significantly increasing overall survival time compared to untreated mice. Protection from ECM by oAC was associated with a reduced numbers of splenic TNF+ CD4+ T cells and multifunctional IFNy+TNF+ CD4+ and CD8+ T cells. Furthermore, we identified a whole blood gene expression signature (68 genes) associated with protection from ECM. To evaluate whether oAC might affect current best available anti-malarial treatment, a group of female C57BL/6 mice infected with PbA received Activated Charcoal. At day 6 p.i., and prior to the first deaths of untreated PbA-infected mice, 5 untreated PbA-infected mice (Group 1) and 5 AC-treated PbA-infected mice (Group 2) were killed and fresh blood (300-500�l per mouse) was collected and processed into RNA. Blood was also taken from uninfected control mice (�baseline�- Group 3).

Digestion of a variety of synthetic peptides by proteasomes

Project description:Proteasomes are large multi-subunit enzymes that act as the main producers of antigenic peptides presented at the cell surface to CD8+ T cells. They can simply cut proteins or recombine their fragments thereby generating novel sequences, i.e. spliced peptides via a process called proteasome-catalysed peptide splicing (PCPS). In order to gain more insight into these processes, we here perform in vitro digestions of 55 synthetic polypeptide substrates by proteasome isoforms followed by LC-MS/MS measurements using different Mass Spectrometers.

Regulation of total and polysomal RNA expression in rat neonatal cardiac myocytes by H2O2

Project description:We are determining the effects of oxidative stress on mRNA expression in cardiac myocytes to identify immediate early genes and establish if there is an effect of different concentrations on recruitment to polysomes for translation. Rat neonatal cardiac myocytes in primary culture were exposed to 0.2 mM H2O2 for 1 h and total or polysomal RNA prepared for microarray analysis. These samples were prepared simultaneously with others for controls, endothelin-1 and insulin that have been published (PMID: 18275597; PMID: 20509958) and these data are already publicly available (E-MIMR-681 for ET-1 data; E-MEXP-2527 for insulin data).
