RBPMS knockdown and overexpression in rat PAC1 pulmonary artery smooth muscle cells (SMCs)

ABSTRACT: Investigation of RBPMS role in post-transcriptional control of mRNAs in rat PAC1 pulmonary artery smooth muscle cells (SMCs). PolyA mRNA-Seq was carried out after RBPMS knockdown in differentiated PAC1 cells and after inducible RBPMS-A overexpression in dedifferentiated (proliferative) PAC1 cells.

Project description:LPS-stimulated macrophages from Pac1+/+ and Pac1-/- mice were compared to assess gene expression changes in the absence of PAC-1 (also known as Dual specificity phosphatase 2). Experiment Overall Design: Age matched Pac1+/+ and Pac1-/- littermates were used to obtain equal numbers of thiolycollate-elicited macrophages. Pure populations of macrophages were stimulated with 100 ng/ml of LPS for 6 hours. 6 hours was chosen to assess mainly inflammatory gene expression.

Project description:RBPMS regulates alternative splicing decision via recruiting other trans factors. This experiment aims to investigate the interactome of Strep-II tagged recombinant RBPMS in Hela nuclear extract. The interactome of the full length protein is compared to the construct lacking the far C-terminal 20 amino acids. The “R” series indicates full-length RBPMS pull-down. The “P” series indicates truncated RBPMS pull-down. The “BL” series indicates full length RBPMS pull-down in the absence of nuclear extract input but with bovine serum albumin beads blocking reagent. The “N” series indicates non-baited negative control pull-down.

Smooth Muscle Cell Phenotypic Switch Induced by Cigarette Smoke Condensate

Project description:Cigarette smoke is a risk factor for inflammatory diseases, such as atherosclerosis. Tobacco smoke interacts with inflammatory cytokines to produce endothelial dysfunction and induces pro-inflammatory and pro-atherosclerotic effects in vascular tissue. Smooth muscle cells (SMCs) are present in the media of human arteries, and are considered protective against atherosclerotic plaque destabilization. Contractile SMCs are the most prominent cell type in the healthy vessel wall. SMCs are not terminally differentiated, and retain the ability to undergo a phenotypic switch from a contractile to a dedifferentiated synthetic state to express inflammatory markers and a phagocytic activity in response to environmental cues. The aim of our study was to evaluate the effects in human SMCs of lipophilic component from cigarette smoke condensate (CSC) and of hydrophilic components of Electronic-cigarette, Tobacco heating products, or cigarette smoke.

Splicing regulatory network of master regulator RNA Binding Protein Multiple Splicing (RBPMS) in human Embryonic stem cell derived Vascular Smooth Muscle Cells.

Project description:We report the splicing and gene expression regulation by RBPMS In hES-VSMCs. We have used a Doxycycline inducible over-expression system to examine the splicing networks controlled by RBPMS in VSMCs; a protein that is highly expressed in native arterial tissue VSMCs. Previously, we have described its splicing regulatory functions in rat VSMCs (Nakagaki-SIlva et al, 2019). Here we report human VSMC specific interactions using VSMCs differentiated from human ES cells via a neural crest lineage. These cells do not endogenously express RBPMS; however, over-expression drives splicing patterns characteristic of mature tissue VSMCs evenin the in vitro differentiated cells.
