Project description:Obtaining adipose tissue samples are paramount to the understanding of human obesity. We have examined the impact of needle-aspirated and surgical biopsy techniques on the study of subcutaneous adipose tissue (scAT) gene expression in both obese and lean subjects. Biopsy sampling methods have a significant impact on data interpretation and revealed that gene expression profiles derived from surgical tissue biopsies better capture the significant changes in molecular pathways associated with obesity. We hypothesize that this is because needle biopsies do not aspirate the fibrotic fraction of scAT; which subsequently results in an under-representation of the inflammatory and metabolic changes that coincide with obesity. This analysis revealed that the biopsy technique influences the gene expression underlying the biological themes commonly discussed in obesity (e.g. inflammation, extracellular matrix, metabolism, etc), and is therefore a caveat to consider when designing microarray experiments. These results have crucial implications for the clinical and physiopathological understanding of human obesity and therapeutic approaches. Project description:Adipose, once considered an inert storage depot, is now known to be an active endocrine tissue involved in total body homeostasis and metabolism, which exerts effects on multiple systems including food intake, immune function, and blood glucose regulation. Adipose tissue depots are known to have unique metabolic and gene expression profiles in vivo and when cultured in vitro. Differences in adipose tissue depot function could be important in determining chronic disease risk. Few comparisons of depot gene expression have been performed in the dog. Utilizing microarray technology, our objective was to identify differentially expressed genes and enriched functional pathways between subcutaneous and gonadal adipose of lean and obese dogs. Subcutaneous and gonadal adipose tissue samples were collected from 9 intact female beagles (4 yr-old; 4 lean controls; 5 obese ad libitum-fed) after 24 wk of ad libitum feeding.

Project description:The aim of the project was to compare global gene expression in adipocytes from obese patients and lean controls. Subcutaneous adipose tissue was collected from severely obese patients undergoing bariatric surgery (average body-mass index (BMI) of 45.5 kg/m2 (n = 12, thereof 4 men) and healthy lean patients undergoing hernia repairs (average BMI of 24.2 kg/m2 (n = 12, thereof 7 men), between 27 and 56 years of age. Adipocytes were isolated by collagenase treatment of adipose tissue, followed by filtering and centrifugation. Floating adipocytes were lysed in Qiazol before RNA purification and microarray analysis.

Project description:Adipose tissue stromal cells contribute to the regulation of adipose tissue in lean and obese states. Myeloid cells such as adipose tissue macrophages (ATMs) and dendritic cells (ATDCs) undergo both quantitative and qualitative changes with obesity. Due to similarity in markers the identify of adipose tissue dendritic cells and macrophages has been elusive. We have refined prior protocols to unambiguously discern ATM and ATDC in mice. We used microarrays to compare the profiles of ATMs and ATDC from gonadal adipose tissue from lean, obese, and formerly obese mice. We also isolated preadipocytes (PA) from lean and obese mice for comparison. Male C57Bl/6 mice were fed normal diet (ND) or high fat diet (HFD) for 16 weeks. Weight loss (WL) mice were switched from the HFD to ND for 8 weeks. RNA was purified from FACS sorted cell populations (live cells only) obtained from gonadal/epididymal adipose tissue depots. ATMs were defined as CD11c+ (CD45+CD64+ CD11c+) or CD11c- (CD45+CD64+ CD11c-) ATMs. ATDC were defined as CD64- CD11c+. PA were defined as CD31- CD45- Sca1+ PDGFRA+.

Project description:Type 2 diabetes is associated with obesity, and is characterized by insulin resistance in target tissues of the hormone combined with insufficient systemic insulin. Insulin resistance apparently begins in subcutaneous adipocytes that fail to further accumulate triacylglycerol. To understand the pathogenesis of transition from lean to obesity and to diabetes, we performed transcriptome profiling by RNA-sequencing of isolated primary human adipocytes. Half of identified transcripts were protein-coding and shared between individuals, while non-protein-coding transcripts were subject specific. Differential gene expression between lean and diabetes reflected corresponding changes in 7 of 12 characterized insulin-signaling proteins. In the tissue, adipocytes are surrounded by extracellular matrix (ECM) providing a dynamic scaffold. However, excess ECM is implicated in obesity and diabetes. Bioinformatic analyses of transcriptomes revealed major upregulation of genes related to ECM in obese compared with lean, which was reversed in diabetes. However, biochemical analysis of adipose tissue demonstrated reduced ECM in obesity or diabetes compared with lean. We conclude that marked changes in the adipocyte transcriptome related to ECM do not reflect global adipose tissue fibrosis, but likely control the pericellular milieu surrounding the adipocyte; and excess ECM is not a feature of subcutaneous adipose tissue in obesity or diabetes.
