Project description:Pituitary belongs to a most important endocrine glands which takes part in the regulation of reproductive functions. The proper functioning of this intermediary between the central nervous system and the target tissues of the reproductive system ensures the proper course of the estrous cycle and affects the female's reproductive potential. It is believed that visfatin, a hormone belonging to the adipokine family, may be responsible for the control of reproductive functions in response to the actual metabolic state of female. Herein we verified hypothesis assuming the modulatory effect of visfatin on the anterior pituitary transcriptome during the mid-luteal phase of the oestrous cycle. We analyzed samples obtained from 5 individuals (n=5). After the enzymatic tissue digestion with the use collagenase V and pancreatin cells were divided for two groups: controls [without treatment] and visfatin (100 ng/mL) treated ones. In vitro cell cultures were conducted for 24 hours.

Project description:BackgroundThe pituitary belongs to the most important endocrine glands involved in regulating reproductive functions. The proper functioning of this gland ensures the undisturbed course of the oestrous cycle and affects the female's reproductive potential. It is believed that visfatin, a hormone belonging to the adipokine family, may regulate reproductive functions in response to the female's metabolic state. Herein we verified the hypothesis that suggests a modulatory effect of visfatin on the anterior pituitary transcriptome during the mid-luteal phase of the oestrous cycle.ResultsRNA-seq analysis of the porcine anterior pituitary cells revealed changes in the expression of 202 genes (95 up-regulated and 107 down-regulated in the presence of visfatin, when compared to the non-treated controls), assigned to 318 gene ontology terms. We revealed changes in the frequency of alternative splicing events (235 cases), as well as long noncoding RNA expression (79 cases) in the presence of the adipokine. The identified genes were associated, among others, with reproductive system development, epithelial cell proliferation, positive regulation of cell development, gland morphogenesis and cell chemotaxis.ConclusionsThe obtained results indicate a modulatory influence of visfatin on the regulation of the porcine transcriptome and, in consequence, pituitary physiology during the mid-luteal phase of the oestrous cycle.

Proteomic analysis of human anterior pituitary gland

Project description:The regulation of pituitary function via the hypothalamus and via intra-pituitary connections represents a complex system. Though hormones secreted from the pituitary glands have been well studied, overall information of proteins expressed in the pituitary glands is very limited. Protein expression profiling of normal pituitary tissue may lead to discovery of novel proteins playing an important role in the physiology of pituitary glands and can lead to better understanding of pituitary gland diseases. We aimed to carry out systematic proteomic profiling of adenohypophysis from human pituitary glands using high-resolution Fourier transform mass spectrometer. A total of 2,175 proteins were identified in this study of which, 105 proteins were identified for the first time as compared to high throughput proteomic-based studies from human pituitary glands. The comprehensive list of proteins identified in this study will facilitate the better understanding the role of this important gland in health and disease.

Proteomic profiling of anterior pituitary of heifer calves offered either a high or moderate plane of nutrition in early life

Project description:Enhanced nutrition during the early calfhood period has been shown to hasten the onset of puberty in heifer calves. The hypothalamic-pituitary-ovarian biochemical signalling axis regulates reproductive development in heifers, with the dynamics of gonadotropin pulsatility within the anterior pituitary gland, in particular, central to final sexual maturity. However, the precise molecular mechanisms regulating the influence of metabolic status on this signalling axis in heifer calves is yet to be fully elucidated. The objective of this study was to determine the effect of an enhanced plane of nutrition during early life, up to 21 weeks of age, on the anterior pituitary gland proteome of heifer calves. Transcription profiling of rat anterior pituitary gland using DES-treated COPxACI F2 rats - Expression QTL mapping.

Project description:Anterior pituitary glands were isolated from 21 week old male rats treated for 12 weeks with the synthetic estrogen diethylstilbestrol (DES). 43 COPxACI F2 animals were selected based on pituitary weight phenotypic extremes: 22 highest and 21 lowest pituitary weights in our F2 population. Expression profiles were determined using Affymetrix Rat Genome 230 v. 2.0 arrays. In conjunction with genomewide genotypes available for these 43 animals, linkage analysis was done using mRNA abundance of each transcript on the array as a separate quantitative phenotype. These arrays provide a resource for expression QTL (eQTL) mapping on a genomewide level in the pituitary gland. Experiment Overall Design: 43 samples from anterior pituitary gland were analyzed. COPxACI F2 male animals were treated with DES for 12 weeks, starting at 9 weeks of age. Pituitary glands were harvested and weighed at sacrifice (21 weeks of age). The 43 samples were selected for pituitary weight phenotypic extremes: 22 highest and 21 lowest pituitary weights in our F2 population. Genomewide genotypes are available for these animals and in conjunction with the array data, genomewide expression QTL (eQTL) mapping was done.
