Transcription profiling of mouse pancreas from Mist knockouts vs. wild type treated withcaerulein (induces pancreatitis) or saline controls

ABSTRACT: Mouse pancreas from wild type and MistKO animals were induced either with caerulein or saline as control and processed for RNA. Targets from three biological replicates of each were generated and the expression profiles were determined using Affymetrix Mouse Expression chips 430. Comparisons between the sample groups allow the identification of genes with differential expression patterns of genes which might contribute to pancreatitis.

Project description:Temporal expression profiling was utilized to define transcriptional regulatory pathways in vivo in a mouse muscle regeneration model. Potential downstream targets of MyoD were identified by temporal expression, promoter data base mining, and gel shift assays; Slug and calpain 6 were identified as novel MyoD targets. Slug, a member of the snail/slug family of zinc finger transcriptional repressors critical for mesoderm/ectoderm development, was further shown to be a downstream target by using promoter/reporter constructs and demonstration of defective muscle regeneration in Slug null mice.

Gene-expression profiling of SEPN1 knockout or wild type mice injected with either ERO1 or PBS

Project description:Endoplasmic reticulum (ER) and oxidative stress are two related phenomena that have important metabolic consequences. As many skeletal muscle diseases are triggered by oxidative stress, we explored the chain of events linking a hyper oxidized ER (which causes ER and oxidative stress) with skeletal muscle dysfunction. Using exon expression arrays we show that the combined genetic modulation of the two master ER redox proteins, selenoprotein N (SEPN1) and endoplasmic oxidoreductin 1 (ERO1), leads to an SEPN1-related myopathic phenotype due to excessive signalling of transforming growth factor (TGF)-beta. One-month-old WT and SEPN1 KO mice were intramuscularly injected with AAV2/1-ERO1a (an adeno-associated virus driving ERO1a) in three sites of the right gastrocnemius muscle and vehicle alone was injected into the contralateral muscle. The animals were sacrificed four-six weeks after injection and total RNA was isolated from muscle tissues using the RNeasy Mini Kit (Qiagen) according to the manufacturer's instructions. Genechip Mouse Gene 2.1 ST arrays (Affymetrix) were used for whole genome gene-expression analysis of duplicated samples derived from WT and SEPN1 KO mice in presence of either ERO1 or PBS.

They were then placed in a normal light-dark environment for 4 days, after which they were sacrificed (at P46). Visual cortex was dissected from 1-mm thick coronal sections, and total RNA was extracted from the tissue lysate using Trizol (Gibco-BRL). Experiment Overall Design: Gene expression in pooled samples from 2 non-deprived (ipsilateral to monocular enucleation) and 2 deprived (contralateral to monocular enucleation) visual hemispheres was compared. Note: two additional cross-animal comparisons were made.
