T. gondii SPARK depletion phosphoproteome

ABSTRACT: Apicomplexan parasites are ubiquitous pathogens of humans. Toxoplasma gondii, a model apicomplexan, possesses a divergent ortholog of mammalian PDK1. This ortholog, TgSPARK, is essential for the ability of parasites to invade and exit their host. Here, we conduct a quantitative mass spectrometry experiment to measure changes to the parasite phosphoproteome following 0, 3, 8, and 24 hours of conditional SPARK depletion.

Project description:Apicomplexans are ubiquitous parasites of humans and cause diseases including toxoplasmosis, malaria, and cryptosporidiosis. Protein kinase G (PKG) possesses unique features in this phylum of parasites and is a demonstrated drug target. Phosphodiesterase inhibitors increase the concentration of cGMP in cells, thereby activating PKG. Here, we generate a phosphoproteome from parasites treated with the phosphodiesterase inhibitor zaprinast. We additionally use an auxin-inducible degron system to deplete parasites of PKG during this treatment.

De novo mapping of the apicomplexan Ca2+-responsive phosphoproteome

Project description:Apicomplexan parasites cause persistent mortality and morbidity worldwide through diseases including malaria, toxoplasmosis, and cryptosporidiosis. Ca2+ signaling pathways have been repurposed in these eukaryotic pathogens to regulate parasite-specific cellular processes governing the transition between the replicative and lytic phases of the infectious cycle. Despite the presence of conserved Ca2+-responsive proteins, little is known about how specific signaling elements interact to impact pathogenesis. We mapped the Ca2+-responsive proteome of the model apicomplexan T. gondii via time-resolved phosphoproteomics. The waves of phosphoregulation following PKG activation and stimulated Ca2+ release corroborate known physiological changes but identify specific molecular targets in these pathways. We characterized protein phosphatase 1 (PP1) as a Ca2+-responsive enzyme that relocalized to the parasite apex upon Ca2+ store release. Conditional depletion of PP1 revealed that the phosphatase regulates Ca2+ uptake to promote parasite motility. PP1 may thus be partly responsible for Ca2+-regulated serine/threonine phosphatase activity in apicomplexan parasites.

Thermal profiling of parasite extracts identified many expected Ca2+-responsive proteins, such as parasite Ca2+-dependent protein kinases. Our approach also identified numerous Ca2+-responsive proteins that are not predicted to bind Ca2+, yet are critical components of the parasite signaling network. Proximity labeling of TurboID-SPARKEL

Project description:Toxoplasma gondii is a ubiquitous parasite of animals. We have previously shown that T. gondii possesses an AGC kinase, SPARK, that is necessary for calcium mobilization, invasion, and egress from host cells. Interaction studies have revealed that SPARK complexes with an elongin-like protein, which we term SPARKEL. Here, we performed proximity labeling of parasites expressing TurboID-SPARKEL endogenously.
